Abstract


In this study, tissue blocks separated adherent method , cultured bovine mammary epithelial cells ; detect the survival of cultured cells , the biological characteristics of the cell growth curve , cell chromosome analysis ; utilize keratin K14 and K15 fluorescence immunoassay staining culture mammary epithelial cells were identified . Followed by liposomal transfection method containing the target gene lacS mammary specific expression vector into bovine mammary epithelial cells . Transiently transfected identification, specific expression vector was transfected into bovine mammary epithelial cells in the most appropriate conditions to determine the breast , and the the transgenic positive cells were identified in the DNA, RNA level . The results show that : containing the gene lacS , breast -specific expression of the carrier can be successfully transfected mammary epithelial cells, and the specific expression . The above research laid the necessary foundation for the subsequent transgenic animal production research .
