This experiment has chosen the pHiladelpHia coronarius ’Aureus’ and Sambucus racemosa ’Plumosa aurea’,its young stems with axillary(stem tips including),as experimental materials to systematically research these works such as aseptic’s gain,start training,subculture proliferation,rooting cultivation and transplant acclimatization.Study the vitro propagation methods of pHiladelpHia coronarius ’Aureus’ and Sambucus racemosa ’Plumosa aurea’.The results are as follow:1.The best sterilization way of explants of pHiladelpHia coronarius Aureus’:use 75% alcohol soaking 30s,wash 3-5 times by sterile water,and 0.1%HgCl2+tween(1-2 drops) sterilizing 6min,and then wash 5 times by sterile water,then the contamination rate is 21.11%.tThe best sterilization way of explants Sambucus racemosa’Plumosa aurea’:use 75% alcohol soaking 10s,wash 5 times by sterile water,and 0.1%HgCl2+tween(1-2 drops) sterilizing 2min,and then wash 5 times by sterile water,then the contamination rate is 22.22%.2.The best startup basic culture medium is N6 for pHiladelpHia coronarius ’Aureus’,Nitsch for Sambucus racemosa ’Plumosa aurea’,the startup rate are 90.00% and 93.33%.The best auxin is IAA,at the same time the best mitogen is 6-BA.3.The best breeding basic culture medium is MS,the multiplication coefficient is 3.09 and 8.02.The best proliferation hormone ratio is NAA0.05mg/L+6-BA0.5 mg/L for pHiladelpHia coronarius ’Aureus’,and NAA0.2mg/L+6-BA2.0 mg/L for Sambucus racemosa ’Plumosa aurea’.4.It is better for their proliferation culture to add sucrose in their proliferation culture medium,the best concentration is 30g/L.The best water quality is distilled water and the best pH is 5.8.When it talks to culture conditions,pHiladelpHus coronarius ’Aureus’ is fit for training room,as contract,Sambucus racemosa’Plumosa aurea’is not strict, training room and climatic cabinate are both allowed.5.The best rooting culture medium is 1/2MS for pHiladelpHia coronarius ’Aureus’,and N6 for Sambucus racemosa’Plumosa aurea’.The rooting rate are 80% and 84.44%.The best auxin are NAA and IBA,with the best concentration 0.5mg/L and 0.6mg/L.The rooting ratio is 95.56% for pHiladelpHia coronarius ’Aureus’ without activated carbon;it is 93.33% for Sambucus racemosa ’Plumosa aurea’ with 2 mg/L activated carbon.The max number of per bottle are 15 and 12.6.Choose the humus,turf and sandy soil as transplant matrix for screening objects.It has been found that the best transplant matrix is humus,the survival rate of pHiladelpHia coronarius ’Aureus’ is 93.33%,and 96.67% for Sambucus racemosa ’Plumosa aurea’.The best way of acclimatization of them are in culture room with the bottle lip open for 2-3days.
