Cloning and Preliminary Function Analysis of a Female Gamete Growth Related Gene FGA1 in Rice

We isolated a reproductive growth related gene, FGA1(female gametogenesis abortion 1), from a T-DNA insertional population of indica rice. The transgenic plants carrying a T-DNA insertion in the gene FGA1 displayed a phenomena that hygromycin resistance and sensitivity segregation ratios showed 1:1 byχ2-test. And the fga1/fga1 homozygous progeny was not found in 6 successive generations. Pollen fertility of fga1 mutant is normal, but the decline of ripening rate and the genotype of FGA1/fga1 exhibited the co-separetion according to the statistical analysis of ripening rate. Reciprocal crosses between FGA1/fga1 heterozygous plants and the wild types showed that the fga1 allele could not be transmitted through the female. All of these indicate that the mutant fga1 is a gametophyte ovary sterile mutant caused by T-DNA insertion.Cytologic observation showed us that pollen fertility of fga1 was as normal as the wild type minghui 86, but the development of female gamete was blocked. In the female gamete of fga1 mutant, we found , egg cell(E), synergids (S), antipodal cells and polar nuclei(P) appeared disaggregation one by one during the 8 nucleuses period, and the fertilization halted. Southern-blot and analysis of the DNA sequence flanking T-DNA showed that fga1 mutant carried one T-DNA insertion at code area of FGA1, which was located on chromosome 5. Above results fully explain that FGA1 are insertional mutants, and the infertility phenotype is not caused by the structure of T-DNA.The consequence of semi-quantitative reverse transcription-PCR indicated that FGA1 expressed during reproductive growth, so FGA1P::GUS was constructed to verify this result combined with GUS. FGA1 gene was isolated to construct expression vectors combined with pCXUN. Transgenic rice plants of the constructs were transform to rice mediated by agrobacterium. These results show that FGA1 is necessary for female gamete maturation. It may form a functional complex and regulate some gene or approach, which plays essential roles during the late stage of ovary formation.
