Abstract


Growing China's aging population , the incidence of prostate cancer in China 's increasing year by year , so the molecular mechanism study of prostate cancer , and to find effective targeted therapies is of great significance . Eph receptors belong to the receptor tyrosine kinase family , and is the largest branch , EphA3 belongs to the EphA receptor . Recent studies have found that the EphA3 cancer development closely related . EphA3 molecules in all human tissues , prostate highest expression level , it is related with the degree of malignancy of prostate cancer is a cancer is an important factor . The SGEF belongs to ephexin family , a SH3 domain of the guanosine exchange factors ( Rho - GEF ) . The study found that the use of RT-PCR and Western blotting methods in the LNCaP cells , PC - 1 can be found in the RNA and protein levels induced by EphA3 gene expression are raised , leaving SGEF expression amount of the RNA and protein levels in a downward trend . Co - IP experiments , we confirmed that there is interaction between the EphA3 SGEF molecules . The interaction between the two EphA3 phosphorylation , and ephrin -A5 - induced strengthening . Thus GST pull down experiments further validate the interaction the EphA3 SGEF N side 216-420 sections and PS section . Construction of the fluorescence expression vector , transfection into C4-2 cells , found EphA3 SGEF molecules are positioned in the cytoplasm , and the two co-transfected cytoplasm after partial colocalization . C4-2 cells of EphA3 with the SGEF high expression to join ephrin - A5 -induced MTT results showed that activation of ephrin - A5 EphA3 little effect on cell growth , the scratch test that may affect cell migration and invasion . We believe that ephrin - A5 may be activated EphA3 phosphorylation , which in turn interact with SGEF activate Rho G. . Thus , we believe that may be SGEF EphA3 Preparation molecular EphA3 by activate SGEF . Thereby activating a series of signaling molecules downstream of Rho G , and thus have an impact on the occurrence of prostate cancer development .
