Objective: Esophageal cancer is one of the common malignant tumor in China, the mortality rate in the fourth place in the malignant tumor. Surgery, radiation therapy, chemotherapy is currently the main treatment for esophageal cancer. As the main means of radiotherapy in patients with esophageal cancer without surgical indications exact. Radiotherapy-induced radiation reactions and radiation damage, limit the amount of radiation therapy of esophageal cancer target affect the efficacy. If you can find to kill tumor tissue in which to the greatest extent, while the surrounding normal tissue damage minimal new radiotherapy method, be greater clinical significance. The Chronobiology Research found, normal tissue and tumor tissue of organisms, each with its own rhythm, and implementation of radiotherapy according to the rhythm characteristics to improve clinical outcomes, reduce radiation reactions and radiation injury provides a theoretical basis. This study to detect esophageal carcinoma cell line of circadian gene hPer1 and hPer2 when phase fluctuations, and then find out the differences in the radiosensitivity of esophageal cancer cell lines of different time points, and expectations for the time of clinical radiotherapy provide some theoretical basis. Method: (1) the in vitro culture of well-differentiated esophageal carcinoma cell line Eca-109. (2) dark to esophageal the cell lines hPer1 rhythm gene expression induced by the nutmeg phorbol ester (PMA), acetyl drugs stimulate esophageal the cell lines hPer2 rhythm gene expression. Conclusion: esophageal cell lines hPer1, low to hPer2 circadian gene expression in 6MV-X-ray irradiation, the higher rate of apoptosis of esophageal carcinoma cell line, the esophageal cancer cells more sensitive to radiation, suggesting that the radiation therapy of esophageal time may be more advantages.
