Abstract


Objective:To investigae the expression of ETS-1and U-PA in non-small cell lung cancer and the their relationship between clnical pathological Parameters, and the relationship between ETS-1and U-PA.To explore their roles in tumorigenesis and progression of non-small cell lung cancer.Methods: Immunohistochemisrty SP mehtod was used to detemine expression of ETS-1and U-PA in 50 specimens of non-small cell lung cancer and 20 specimens of normal tissues, then investigae their relationship between clnical pathological parameters, and the relationship between ETS-1and U-PA. The expression of Ets-1 and u-PA mRNA were detected by reverse transcriptase polymerase chain reaction (RT-PCR) assay on 17 specimens of NSCLC and corresponding normal tissues, to explore the relative expression intensity of Ets-1 and u-PA mRNA in non-small cell lung cancer and corresponding of normal tissues.Results:①The expression rate of Ets-1 and u-PA protein in NSCLC tissue were 70%(35/50) and 64%(32/50), which is significantly higher than that in the normal tissues, 15%(3/20)and 10%(2/20)(P<0.001), respectively.②The expression rate of Ets-1 protein was increased in deeper of invasion, lymph node metastasis and higher clinical stages (P<0.05) of NSCLC, but was not associated with sex, age, smoking, histological classification and differentiation (P>0.05).③The expression rate of u-PA protein was increased in lymph node metastasis and higher clinical stages (P<0.05) of NSCLC, but did not associate with sex, age, smoking, histological classification, differentiation and depth of invasion (P>0.05).④A positive correlation between the expression of Ets-1 and u-PA protein was found (rs=0.452, P<0.01).⑤The relative expression intensity of Ets-1and u-PA mRNA in 17 NSCLC tissues were 0.564±0.059 and 0.773±0.191, significantly higher than that in corresponding normal tissues (0.318±0.089 (P=0.000), and 0.401±0.193 (P=0.000)).Conclusion: Over-expression of Ets-1 and u-PA correlate closely with carcinogenesis and progression of NSCLC. There was a positive correlation between the expression of Ets-1 and u-PA protein. Ets-1 might be involved in metastasis of NSCLC via up-regulateing the expression of u-PA .
