Objective To study the Guangzhou region carrying bla CTX-M-15 plasmid molecular characteristics. 181 the period from 2007 to 2008 from the nine hospitals in Guangzhou ESBL-producing E. coli and 180 K. pneumoniae bacteria, the PCR method of screening for carrying the bla CTX-M-15 strains constitute determine the resistance gene of Escherichia coli phylogenetic groups and analysis of popular plasmid PFGE and MLST about bla CTX-M-15 -positive strains homology, MIC detection of strains commonly used antibiotics. The serum agglutination test to determine the serotypes. Bonding experiment understanding carry BLA CTX-M-15 plasmids can bondability, restriction enzyme analysis of plasmid homology. The second generation sequencing technology plasmid sequences were sequenced. The results detected bla CTX-M 243 positive strains, CTX-M-14 and CTX-M-15, respectively, 115 and 84. 37 Escherichia coli and 47 Klebsiella pneumoniae carrying bla CTX-M-15 . bla CTX-M-15 -positive Escherichia coli and Klebsiella pneumoniae were used PFGE identified as 28 and 30 genotypes. Carrying bla CTX-M-15 Escherichia coli phylogenetic group D group (67.8%) and group A (21.4%). Serotypes of E. coli was not found 025: H4 serotype scattered. ST-type of Escherichia coli and Klebsiella pneumoniae no obvious central tendency. 58 representative strains of genotype 40 carrying bla CTX-M-15 plasmid passed to the recipient strain E.coli C600 (Rifr), Escherichia coli 19 Klebsiella pneumoniae 21. 35.7% (10/28) of Escherichia coli carrying bla CTX-M-15 plasmid size to 65KB, 40.0% (12/30) of Klebsiella pneumoniae carrying bla < sub> CTX-M-15 plasmid size is 92KB. By restriction endonuclease analysis, and found that there are two popular plasmid, named p 15-e sup> and p 15-k sup>. PCR survey the p 15-e sup> exist the bla CTX-M-15 and ISEcpI; bla p 15-k sup> CTX-M-15 , blaTEM-1. The analysis the p- 15-k sup> full sequence found the plasmid found with Zhejiang pKF3-94 has significant homology. Conclusion the Guangzhou region CTX-M-type ESBL CTX-M-14; CTX-M-15 came in second place, but the upward trend is very clear. Guangzhou regions carrying bla CTX-M-15 strains independently genotype-based, there is no evidence of large-scale spread of bacterial clones. Carrying bla CTX-M-15 of the Escherichia coli phylogenetic group to group D and group A mainly with foreign countries there are regional differences. Guangzhou area carrying bla CTX-M-15 Escherichia coli and Klebsiella pneumoniae ST and serotype differences are reported abroad. bla CTX-M-15 exists in the bonding properties of grain on. E. coli is mainly about the size of 65Kb plasmid; the Klebsiella pneumoniae focused 92Kb plasmid. Guangzhou region of Escherichia coli and Klebsiella pneumoniae were popular a 65Kb (pc15-e) and 92KB (pc15-k) plasmid. Plasmid pc15-k and Zhejiang found pKF3-94 having a high degree of homology, suggesting that The Chinese domestic there may be the one carrying the bla CTX-M-15 Pop plasmid.
