The research of Quercetin 2,3-dioxygenase is a newly emerging hot topict in bioinorganic field. Based on the research of 2,3-QD active site structure and dioxygen activation, to further clarify the catalytic role of Glu73, metal ion effects, electronic effects and steric effects on the structure and reactivity and activation mechanism of dioxygen of 2,3-QD, we design, synthesize and characterize a series of structural and functional model complexes, and study the reactivity towards flavonol and dioxygen of the model comlpexes, the details are as follows:One new 2-((bis((6-chloropyridin-2-yl)methyl)amino)methyl)benzoic acid (L12H) has been designed, synthesized and characterized by MS, IR, 1HNMR.Six novel binary model complexes and Six ternary model complexes M(Ⅱ) (M:Mn, Fe, Co, Ni, Cu and Zn) have been designed, synthesized, and characterized as the structural and functional model complexes for the active site of 2,3-QD. Our design of model ligand and complexes focus on both catalytic site and substrate binding site of the enzyme.The reactivity of the model complexes towards flavonol and dioxygen have been investigated by UV-Vis monitoring. Our conclusions are as follows.(Ⅰ) The same ligand with different metal ions, the reactivity of model complexes display some differences and in the order of Ni(Ⅱ)>Co(Ⅱ)>Cu(Ⅱ) for binary complexes and Fe(Ⅱ) >Co(Ⅱ)>Ni(Ⅱ)>Zn(Ⅱ)>Mn(Ⅱ)>Cu(Ⅱ) for ternary complexes. Although the structures of the model complexes are similar, the reactivity shows notable differences.(Ⅱ) With the same ligand and metal ion, the reactivity of ternary complexes is much better than corresponding binary complexes.(Ⅲ) The same metal ion with different ligands, the reactivity of L12 (with 6-C1 substituted pyridine ring) model complexes (except copper complexes) is higher than corresponding non-substituted L1 complexes, and in the order of L12>L1. The reactivity of Cu(II) complexes of L1 is higher than L12, maybe it is contributed to the electronic effects and steric effects of the ligand and metal ion effects on the reactivity.
