Abstract

Infertility is one of four disease of dairy cow. The main causes of dairy cow infertility is inflammation of genital tract. Infertility lengthens the ICC(the interval from calving to conception) and decreases milk production and causes serious economic losses. Conventional treatments with antibacterial substances could induce residues in milk, spread of bacterial resistance, increase in financial costs. Therefore, increasing preventive treatments with natural products which would avoid alterations in milk quality and economic losses due to decreased reproduction. Some of these products are probiotics. The main contents of the research are study the microbial flora of dairy cow’s genital tract, isolate and identify lactobacillus from genital tract of healthy cows and study the Lactobacillus biological characteristics of healthy dairy cow’s genital tract, and the following results:1. Collected of ten healthy dairy cows and ten dairy cows with infertility due to genital tract inflammation from dairy farms in Inner Mongolia Hohhot. Isolated and identified bacteria in dairy cow’s genital tract using standard microbiological methods. The most dominant bacteria in genital tract of healthy dairy cows belong to the lactobacillus(P<0.01); The most dominant bacteria in genital tract of infertile disease dairy cows belong to Staphylococcus aureus(P<0.01); The quantity and rate of isolation of lactobacillus and Streptococcus in healthy dairy cows was obviously higher than the infertile disease cows; The quantity and rate of isolation of Staphylococcus aureus in genital tract of infertile disease dairy cows was obviously higher than the healthy dairy cows; Coliform group, Staphylococcus epidermidis and others with no significant difference between the groups(P> 0.05).2. Isolated 55 lacto acid bacteria from genital tract of healthy cows, and 6 lacto acid bacteria of them which have superior growth performance and strong acid production capacity were further identified. The 6 lactic acid bacteria included Lactobacillus plantarum, Lactobacillus brevis, Enterococcus faecalis, Lactococcus garvieae, Lactobacillus kitasatonis, Lactobacillus amylovorus.3. The 6 lactic acid bacteria with superior growth performance and strong acid production capacity were further sdutied. The results show that the number of 12 and 15 and 45 and 49 strains have superior growth performance and strong acid production capacity and have obviously inhibitory effects on Staphylococcus aureus and pathogenic Escherichia coli(P<0.01); The number of 30 strain have ordinary growth property and acid production capacity, and have inhibitory effects on Staphylococcus aureus and Pathogenic Escherichia coli(P<0.05).The number of 41 strain have weak growth performance acid production capacity, and have not inhibitory effects on Staphylococcus aureus and pathogenic Escherichia coli(P>0.05). Therefore, the number of 12 and 15 and 45 and 49 strains can be used as candidates for the preparation of microecological additives of dairy cows and need further study.
