Study on the Lipoprotein Purification with Tara-gum and Chitosan Sulphonated Derivatives

Abstract


In this paper, lipoprotein apheresis agents and adsorbent were synthesized by using tara-gum or chitosan as raw materials, sodium 3-chloro-2-hydroxy propanesulfate or chlorosulfonic acid as sulphonating agents. Their properties for selective purification of total cholesterol(TC), low-density lipoprotein cholesterol(LDL-Ch) and very low-density lipoprotein cholesterol(VLDL-Ch), high-density lipoprotein cholesterol(HDL-Ch), total protein(TP) and fibrinogen(Fib) from plasma were investigated by induced precipitation and adsorption in vitro. The main conclusions were made as follows:1) Degraded tara-gum sulphonates (DTS) was synthesized with tara-gum degraded by H2O2 and sodium 3-chloro-2-hydroxy propanesulfate as ligand. IR spectrum analysis showed that DTS was a novel heparin-like material. The results of lipoprotein apheresis with DTS showed that, under proper conditions, the average purification percentages of TC, HDL-Ch, LDL-Ch+VLDL-Ch, TP and Fib was 47.2%,40.4%,51.3%,12.4% and 94.4% respectively.2) Tara-gum sulphonates (TS) were synthesized with tara-gum as raw material, and chlorosulfonic acid as ligand. IR spectrum analysis showed that TS was a noval heparin-like material. The results of lipoprotein apheresis with TS showed that, under proper conditions, the average purification percentage of TC, HDL-Ch, LDL-Ch+VLDL-Ch, TP and Fib was 53.4%,33.1%,60.1%,11.7% and 94.7% respectively.3) Spheric chitosan sulphonates (CHS) was synthesized with chitosan as carrier, sodium 3-chloro-2-hydroxy propanesulfate as ligand. The results of batch adsorption with CHS under proper conditions showed that the average adsorption percentages of TC, HDL-Ch, LDL-Ch+ VLDL-Ch, TP and Fib was 15.6%,10.4%,18.9%,12.2% and 13.5% respectively; and the continuous adsorption percentage of CHS to TC, HDL-Ch, LDL-Ch+VLDL-Ch was 20.4%, 13.8% and 24.6% respectively, which showed a certain selective adsorption ability of Lipoproteins.The lipoprotein apheresis agents synthesized in this paper possess good properties of selective purification of lipoprotein. This research provides good basis for the further deveopment of lipoprotein purification.
