Background: Atherosclerosis (atherosclerosis, AS) serious harm to human health, it causes heart and cerebrovascular diseases to bring a physical and mental suffering, but also to society brought a heavy economic burden, research The molecular mechanism of the development of AS is the treatment of AS premise and key. In the development of AS many molecular mechanisms, peroxisome proliferator-activated receptor γ (peroxisome proliferators activated receptors, PPARγ) as a class of ligand-activated nuclear receptors, and its role has been fully confirmed. PPARγ can influence lipid balance and change the energy metabolism, regulation of inflammatory responses, and in the formation of foam cells and vascular smooth muscle cells (vascular smooth muscle cell, VSMC) proliferation and collagen secretion play a role, but the specific mechanism is not clear. The TGFβ / smad is another in the development of AS play an important role in the signaling pathway, which through smad pathway to promote VSMC proliferation and collagen secretion. c-Ski as a repressor of the signaling pathway can block the signal transduction pathway, the mechanism is smad3 the MH2 domain binding thereby preventing transcription complex formation and raise corepressor child. These studies suggested c-Ski as background TGFβ / smad pathway repressor of AS may play an important role in the occurrence and development, but on the c-Ski in the AS specific role and whether there is a correlation between PPARγ adjustable contact no report . Clarify this problem, you can clarify the interaction between the two in the AS and mechanisms for the treatment of AS to provide new ideas and strategies. Objective: To determine the AS PPARγ and c-Ski interconnected and influence of the AS. Methods: The rat through a high-fat diet AS model, testing AS plaque c-Ski mRNA and protein expression changes; in cultured rat vascular smooth muscle cells (vascular smooth muscle cell, VSMC) transfected with recombinant c-Ski plasmid and observe its effect on VSMC proliferation and collagen secretion in rats; using real-time PCR and Western blot PPARγ agonists and antagonists on the regulation of expression of c-Ski, and then use the online program NUBIScan and luciferase reporter Genetic testing for c-Ski clearly PPARγ regulation mechanism. Results: (1) AS rat arterial plaque c-Ski mRNA and protein expression was significantly reduced. (2) recombinant c-Ski after plasmid transfection significantly inhibited rat VSMC VSMC proliferation and collagen secretion. (3) PPARγ agonist rosiglitazone significantly increased rat c-Ski expression in VSMC, and this increase could be blocked by PPARγ antagonist GW9662. (4) bioinformatics analysis showed that the promoter region of c-Ski possible PPARγ binding sites, and rosiglitazone also significantly turned on dyed c-Ski promoter reporter gene expression. CONCLUSION: c-Ski effect with certain anti-AS, which may be another PPARγ target genes can activate PPARγ increases its anti c-Ski AS effect.
