Study of Extracting Diosgenin from Dioscorea Zingiberensis Using Surface Active Agent and Ultrasonic Method

Abstract


Objective: defects in the turmeric diosgenin extraction process exists to explore the establishment of a new, efficient extraction process route . Method: ethanol solvent, surfactant combined with ultrasonic method diosgenin extracted from turmeric ; using single factor test and orthogonal test the concentration of ethanol , Soxhlet extraction time , [H] and other conditions were optimized to get the best extraction process conditions ; using UV - visible spectrophotometry by RP-HPLC determination of diosgenin content ; recrystallized from methanol purified diosgenin and experimental characterization of its melting point , IR , TLC . Results: surfactant combination of ultrasonic extraction of diosgenin optimal extraction conditions for the solid-liquid ratio 1:12 , ultrasonic 45min , ultrasonic temperature 65oC, cumene sulfonate concentration of 10mg/mL . Diosgenin average yield of 2.37% , the yield increased by 12.86% compared with traditional direct acid hydrolysis wears , and measured by RP-HPLC method diosgenin purified to a purity of 94.05% . Conclusion: The surfactant combination of ultrasonic method extracted diosgenin yuan yield was significantly higher than traditional direct acid hydrolysis . In addition , methanol recrystallization purification effect of diosgenin than ideal .
