Abstract


Alzheimer's disease (Alzheimer's disease, AD) is a progressive memory loss and cognitive impairment as the main symptoms of neurodegenerative diseases , in two distinct histopathological features of the patient's brain : the intracellular neurofibrillary tangles (neurofibrillary tangle, NFT) and extracellular amyloid plaques ( the senile plaque , SP ) is formed . Studies show SPs contains large amounts of β-amyloid protein (beta-Amyloidprotein, Aβ), BACE1 (β-site APP-cleaving enzyme; APP, amyloid precursor protein) is considered to be a key enzyme of Aβ formed has become Arz Alzheimer research hot spots. But there is still a lack of comprehensive understanding of the physiological functions and mechanisms of action of BACE1 . Our laboratory to BACE1 The intracellular end of the 473 to 501 amino acids in order to recognize and understand more clearly the role of BACE1 in the pathogenesis of Alzheimer's disease process , build a bait protein using the yeast two-hybrid method in human fetal brain cDNA library was screened to obtain a BACE1 interacting proteins : the pp6068 clone protein . We build pp6068 cloned proteins in mammalian cells stably expressing vector , co-immunoprecipitation experiments show that exogenous pp6068 cloned protein BACE1 able to interact in HEK293T cells . N2a/APP695 cells , pp6068 cloning overexpression of upregulated BACE1 hydrolysis product of APP / βCTF and Aβ protein levels , it is also able to cut the α- secretase enzyme hydrolyzate sAPPα protein levels . These studies illustrate , pp6068 cloned protein may be involved in regulating the activity of the BACE1 , influence the formation of the degradation of the APP and Aβ . Proteolytic processing of APP α- secretase enzyme may also affect . For the study of the interaction with BACE1 protein is not only conducive to reveal the biological function of the the pp6068 cloned protein in mammals , may also open new avenues for therapeutic research BACE1 physiological function and AD .
