Elements of Thirty Kinds of Proprietary Chinese Medicines and Classification Based on Elements

Abstract


Objective: The experiment chose commonly used Chinese traditional medicine treating diabetes, stomach diseases and cardiovascular diseases. Detecting and analyzing the element content of them so that we can explore the reference method of classification on these medicines based on element content. The purpose is researching and exploring the correlation between element content with medicine efficacy or classification.Method: Chose thirty kinds of proprietary Chinese medicine samples, and remove the water of them to make into dry powder. Weighed 0.2-0.3g dry samples and placed in the digestion tank. Made a reagent blank as contrast at a same time, then add 5mL nitric acid, standing an hour, and adding 2mL H2O2.Cover with closures and put it into microwave digestion. Put the digestion sample into inductively coupled plasma-mass spectrometry (ICP-MS), and selected 23 elements as variable to classify the medicine. Draw elements map of them and made factor analysis and cluster analysis.Result: The experiment mainly from two aspects of elements and samples: from the perspective of factor analysis we can see that these elements can be divided into two groups. The above group were mainly trace elements, and semi-trace elements Fe56 and Fe57 stand between the two groups, and the below group were mainly major elements. Toxic elements stand mostly around the two groups and relatively far from the center of group. Constructing the elements map of this medicine we find that they are rich in Na/Mg/Al/K/Ca and essential trace elements Zn/Mn/Co/Ni/Mo, etc. The result angle on cluster analysis from the sample point not only show that classification based on element contents’result basically agree with that of Chinese Traditional medicine, but also provide reference method for not elsewhere classifiable medicine. The result prompts from the new view on perspective of elements possible quest a new proprietary classification.Test on harmful elements and radioactive elements show that the content of four harmful elements Pb/Cd/Hg/As in thirty samples mostly coincidence with the Green medicinal plant import draft standards and proprietary standards for imports in Southeast Asia, and still exists gap with Ninth National People“standardization of traditional Chinese medicine quality standards”and the French standards for imported Chinese Herbal Medicine, and exists a wide gap with FDA drug and functional food standards.Tests results from three radioactive elements can be seen thirty kinds of proprietary Chinese medicines in U/Th/Tl content is much lower than the national occupational health standards for toxic doses.Conclusion: Elements content in Chinese traditional medicine treating the same disease has great differences. So constructing elements map can be benefit on the research of Chinese traditional medicine. The same elements in these drugs’distribution are different even widely. Some of essential trace elements are related with efficacy and categories of these medicines. Cluster analysis of trace elements provides reference data for medical classification and efficacy analysis which is characterized by element contents. Clusters came from three categories of medicine have certain fit with Traditional Chinese medicine theory, and this point indicated that building elements map on elements content and then make statistical analysis on elements character may have some kind of significance on medicine identification.
