This thesis focuses on polymerized hemoglobin as a kind of oxygen carriers. Oxygen carriers could be widely used. They can not only apply as blood substitutes, but also use as oxygen therapeutics. Polymerized hemoglobin has been studied for about twenty years. However, it failed to approve clinical trial phase III because of its two main flaws. One is that its complex composition makes its property very uncontrollable. The other is that the product contains a little of unpolymerized hemoglobin which will cause the product very unsafe.My proposal is to solve these two main flaws. I use sulphydryl-cross-linkers such as dimaleimides instead of amino-cross-linkers such as glutaraldehyde to polymerize hemoglobin. This proposal would solve the problems. First, sulphydryl groups (about two) are much less than amine groups (more than forty) on the surface of hemoglobin. Therefore, the process of polymerizing is much easier and more controlable. Sencond, the reaction has a very high activity and is almost quantitative yield. So it will decrease the ratio of unploymerized hemoglobin.By electrophoresis, size exclusion chromatography and other analyses, my work has proved that under pH 9 condition, the product using sulphydryl-cross-linkers such as dimaleimides has easier composition than the product using glutaraldehyde does.
