Schistosoma Japonicum Glucide Metabolic Pathways Based on Bioinformatics Analysis

Few progress has been achieved on the development of vaccine and drugs of the schistosomiasis which afflicting over 200 million people in 76 countries of the world. The glucide metabolic pathways, which are evolutionarily conserved and existed nearly in all organisms, play key roles in cellular processes. There are many potential vaccine targets existed in these pathways. Here, we reconstructed the glucide metabolic pathways in Schistosoma. japonicum and compared the schistosome glucide metabolic pathways with those of model eukaryote species. We totally identified 94 homologous components in the glucide metabolic pathways of S.japoncium and among them, 18 were predicted to be full-length sequences. Phylogenetic analysis of these proteins confirmed the evolutionary conservation of the glucide metabolic pathways. Remarkably, some members we identified in the 5. japonicum glucide metabolism are more original and even could not be found in the Metazoa, these components could be the potential targets for the vaccine and drug devolepments. Further more, we constructed a strategy to optimize the transcriptome database and to get full length coding sequences quickly, 10 groups of partial sequences we identified in the glucide metabolic pathways in the transcriptome database matched to the N terminal and C terminal of the same proteins respectively. Each group was predicted to be assembled into one sequence. RT-PCR was used to close the gaps between them and the new full coding sequences were reassembled. One of them was submitted to GenBank and the Assess Number was EF503564. This strategy is also useful to the optimization of other transciptome database. Therefore, the identification of putative molecular components of schistosome glucide metabolic pathways should lead to a better understanding of fundamental biological process including growth, development and immune escape in the parasite, and likely promote the development of novel vaccine targets and therapeutic interventions effective against schistosomiasis.
