Respiratory syncytial virus (RSV) is the commonest pathogen of the infants with respiratory infection diseases. After RSV infection, the ratio of T helper cell type-1 (Thl) and type-2 (Th2)i(?) unbalanced, and mechanism can be induced by the released cytokines. IL-6, IL-8, IL-12 are no only important cytokines in the lung, but also important roles of Toll-like receptor (TLRs) signa transduction pathway. They are might be involved in the development of RSV infection, and play an important role in the inflammatory response and immune defense. The clinical and experimental researchs show that Jinxin Oral Liquid(JOL) has the effect to resist RSV infection and its mechanism is unclear. It is suggested that JOL may plays the antivirus action by affecting the expression of cytokines. This study is conducted on this basis.Objectives:1.To observe levels of serum IL-6,8,12 in RSV infection, and suggest the mechanism of RSV infection;2.To observe impact of IL-6,8,12 by JOL and to explore mechanism of anti-RSV infection.Methods: With methods of cell culture, laboratory animal science, molecular immunology, ELISA assay we detected the alteration of IL-6,8,12 expression. Data analysis was done with SPSS 16.0.Results:1. After infection,IL-6, IL-8 levels were higher in model group (P<0.05), nevertheless IL-12 levels were significantly decreased (P<0.01);2. After JOL intervention,IL-6,IL-8 levels in high, medium and low dose and pre-treatmen groups (HDG, MDG, LDG,PTG)were significantly lower than those in the model group (P<0.01) otherwise IL-12 levels in HDG,MDG,PTG were significantly increased (P<0.01).3. Compared ribavirin group, the expressions of IL-6 were significantly lower in HDG,PTC (P<0.01); IL-12 levels were significantly higher in HDQMDQPTG.(P<0.01).4. By preintragastric,IL-6 was lower than that by medium,low dose (P<0.01).IL-8 was also lowe by low,medium and high dose. While IL-12 was increased significantly (P<0.01).Conclusions:1.IL-6, IL-8, IL-12 is the important cytokines for RSV infection. After infection,IL-6, IL-8 level: rose, and IL-12 level reduced.2. JOL can play the anti-RSV action. It can reduce the serum IL-6, IL-8 expression, and increase scrum IL-12 expression. This may be one of the mechanisms of anti-RSV action.
