Abstract


Objective: To investigate the the Guangxi crowd HLA-DRB1 * 15 gene and the corresponding sites under Th1/Th2 cell IFN-γ, IL-4 on hepatitis B vaccine immune response levels, clarify the HLA-DRB1 * 15 and IFN-γ, IL -4 relations role in the response to the hepatitis B vaccine, hepatitis B vaccine failed to understand mechanism of immunogenetics. Method: select enrollment to complete a recombinant hepatitis B vaccine standard fully immunized (10μg / 935, 0,1,6), Han nationality in Guangxi healthy college students, aged between the ages of 20-24, extracted detection of hepatitis B virus in peripheral blood serum Markers (HBsAg, anti-HBs, anti-HBc) and liver function. S / N value screened under the anti-HBs titers <10 weak, 80 non-responders, and randomly selected anti-HBs S / N value> 10, the strong response by 62 as the research object. The selected object detection had no past and the present disease HBV infection history, and no history of other chronic diseases, normal liver function, HBV-DNA negative. By PCR-SSP HLA-DRB1 * 15 allele detected by ELISA in serum Th1/Th2 cell factor IFN-γ, IL-4 the OD values ??calculated concentration values ??and comparative analysis. Results: ① weak response group, HLA-DRB1 * 15 gene expression frequency of 11.25%, significantly lower than 36.67%, the strong response group (P = 0.001); ② anti-HBs, weak responder group, IFN-γ The average expression levels (6.28 ± 6.84) pg / ml, significantly lower than the anti-HBs strong response group (16.28 ± 10.05) pg / ml (P = 0.000); weak response of IL-3 anti-HBs No, 4 average expression level as (2.36 ± 1.91) pg / ml, also significantly with less than anti-HBs, strong response group (8.50 ± 6.68) pg / ml (P = 0.000); ④ of HLA-DRB1 * 15 positive expression group IFN -γ in the average expression level for (13.82 ± 9.89) pg / ml, significantly higher than that of HLA-DRB1 * 15-negative expression group (9.76 ± 9.54) pg / ml (P = 0.029); ⑤ of HLA-DRB1 * 15 positive expression group IL -4 average expression levels (5.81 ± 4.27) pg / ml, HLA-DRB1 * 15-negative group IL-4 average (4.82 ± 5.84) pg / ml, no statistically significant difference between the two groups (P = 0.207) . Conclusion: ① The of HLA-DRB1 * 15 gene might is enhanced Guangxi crowd hepatitis B vaccine response of related genes; ② The of Th1/Th2 cell related factor of IFN-γ, of IL-4 the expression level may affect the body hepatitis B vaccine's immune effect; ③ The of HLA-DRB1 * 15 gene may be by affecting the level of associated factor of IFN-γ in Th1 cells generated, thereby enhancing the immune response of the hepatitis B vaccine. ④ HLA-DRB1 * 15 gene may not affect Th2 cell cytokines IL-4 level generation and enhance the immune response of the hepatitis B vaccine.
