Objective To compare Candida albicans CHK mutant CHK26 in (CHK Ser / Thr MAPK and GAF ??domains deficient strain), CHK21 (CHK allelic deletion strain) and standard strain CAF-2 pathogenicity in mice to understand CHK Its domain of the role in the pathogenicity of Candida albicans. Method 1. Establish a mouse model of systemic Candida albicans infection: 18 clean healthy Kunming mice were randomly divided into three groups A, B, C, n = 6, respectively, by tail vein injection of C. albicans CAF-2 , CHK26, and CHK21 overhang the broth 0.5ml (1 × 10 6cfu). Remove the mouse kidney fungal culture do histopathological examination and renal tissue. Mortality in mice, and the survival of protected time experiment: ibid constructed mouse model 63, the morbidity and mortality of the mice in each group (n = 21), twice daily observation. Renal histopathology and electron microscopy detection: to build mouse model 12, n = 4. 3 day and 5 days a mice model kidney, histopathology, PAS staining, another kidney produced electron microscopy specimens were observed under transmission electron microscope. 4. Spleen and kidney tissue colony forming units (cfu): mouse model 45, 15 in each group. 1 day, 3 days and 5 days out made of five mice of both kidneys and spleen homogenates After incubation, count the number of colonies. Compare each group of mice spleen and kidney tissue colony forming unit count differences. 5 standard strains of Candida albicans, CHK the mutant CHK26 and CHK21 preparation electron microscopy and scanning electron microscopy specimens, electron microscopy ultrastructure. Results 1 successfully established a mouse model of systemic Candida albicans infection: three groups of mice kidney histopathology PAS staining visible obvious hyphae and spores, kidney tissue culture to see the growth of yeast-like colonies. Experimental results on mortality and survival protection: standard strain, CHK26 the CHK21 infected mice observed during mortality were 100%, 23.8% and 0 the; CHK21, CHK26 infected mice survival time was significantly longer than the standard strains of mice, statistically significant. 3. The kidney histopathological and electron microscopy test results: kidney tissue pathology detection: the standard the strain, CHK26 and CHK21 infected mice kidney tissue biopsy PAS staining visible fuchsia hyphae and spores. The transmission electron microscope to see: three Candida albicans infection in renal tissue were seen in the epithelial cells of Candida albicans spores. Standard strains of mice infected kidney tissue ultrastructure change significantly, showing tubular epithelial cell surface microvilli shedding even focal necrosis; epithelial cells mitochondrial swelling; renal interstitial neutrophil infiltration ultrastructural changes. CHK26 mice infected kidney tissue lesions lighter, individual tubular epithelial cell edema; lysosomes; renal interstitial infiltration of neutrophils. CHK21 infected mice kidney tissue structure generally normal; only see the individual epithelial cell swelling; small amount of neutrophil infiltration. 4. Spleen and kidney tissue colony forming units (cfu) Results: Candida albicans infection in mouse spleen and kidney tissue colony count results as shown in Table 3: CHK21 clear the slowest in the spleen and kidney tissue, CAF-2 Clear fastest, CHK26 The between standard strains and CHK21. Candida albicans transmission electron microscopy see CHK26 cell wall thickness varying electron-dense outer layer and a transparent inner layer is significantly less than the CAF-2, or even part of the cell wall disappear; basic integrity of the cell membrane. Complete CHK21 cell wall of varying thickness, the thickness in between. The scanning electron microscope, see three Candida albicans round or oval shape, smooth surface; the standard strains CHK26 strains seen in the major axis ends of the cells budding; while the surface of the CHK21 strain cells bud scar random distribution. Conclusion 1. CHK Candida albicans virulence play an important role, CHK21 performance is non-toxic, CHK26 manifested as reduced virulence in a mouse model. Candida albicans infection of a mouse model of renal tissue lesions mainly in the renal tubules. Mice infected with Candida albicans, renal tissue in renal tubular epithelial cells engulf Candida mitochondrial swelling, renal interstitial neutrophil cell infiltration ultrastructural changes mainly. 3. CHK affect Candida albicans cell wall construction, may also be virulence reduce the main reason. 4. Ser / Thr MAPK and GAF ??domains in Candida albicans virulence and cell wall building plays an important role. Changes in the structure of the cell wall, it may affect the budding of Candida.
