Abstract


Part of heterogeneous microorganisms in the intestine due to degradation of the amino acid and many health and harmful substances such as phenol , cresol, indole, secondary bile acids and female hormones, etc. , these harmful substances often become mutagenic agents , in vivo long-term accumulation induced colon carcinogenesis . MRS medium , add the phenol , the cresol and indole potential of the mutagenic substances , investigated the Lactobacillus pentosus food grade Ind1 and Ind3 as well plant Lactobacillus Lp115 clear these mutagens , for prebiotics bacteria in vitro screening and health effects evaluation provide the basis . The results are as follows : (1) in MRS medium was added 0, 5, 50 and 150 μg of phenol, cresol or indole , the the Lactobacillus pentosus strain Ind1 and Ind3 and Lactobacillus bacilli Lp115 tolerated . trials found that concentrations greater than 150 μg of phenol , cresol , and indole substantially no effect on the growth of the strain , 37 ° C anaerobic culture of the number of viable cells after 144h 10 ~ 6cfu/mL . (2) The concentration of phenols in the simulated human intestinal environment , the Lactobacillus pentosus of the Ind1 and Ind3 in cultured under 37 ° C anaerobic conditions 144h , two Lactobacillus pentosus , different concentrations of viable bacteria in different phenols number is still maintained at 10 ~ 6cfu/mL, Lactobacillus pentosus phenols harmful intestinal high tolerance . (3) right MRS medium , cell - cell adhesion as well as degradation of the effectiveness of the mutagenesis factor analysis showed that at concentrations of 5μg/mL and 50μg/mL of phenol and cresol medium , Lactobacillus pentosus the Ind1 and Ind3 Lactobacillus plantarum were cultured at 37 ℃ Lp115 after 24h and 144h , phenol and p-cresol content was significantly reduced, its concentration in 0.95μg/mL 1.93μg/mL between the p-cresol concentration in 0.45μg/mL and 1.14 μg / mL between the number of viable cells of three Lactobacillus from 10 ~ 9cfu/mL to 10 ~ 6cfu/mL, reduce the amount of adsorption of mutagenic factors with the time extension of the cell culture , showed that Lactobacillus cells factor mutations there are good adhesion or degradation . The measurements show that the phenol content in the three Lactobacillus bacterial cells between 0.42μg/mL and 1.47μg/mL cresol content in 0.90μg/mL and 1.28μg/mL between , that Lactobacillus cells with a certain adhesion mutagenesis factor of capacity , but with the incubation time, the decline in the viable cell count of the cells , cell adhesion mutagenic effect decreases .
