Abstract


The biological function of telomeres protect the ends of chromosomes , avoid nuclease degradation of the ends of chromosomes to prevent fusion of chromosome rearrangement , to maintain the integrity and stability of the genome structure plays an important role . Telomerase telomerase reverse transcriptase ( TERT ) and telomerase RNA template ( TRC ) has a special reverse transcriptase activity in ribonucleoprotein complexes. PinX1 is a conserved nucleolar protein interaction with the telomere / telomerase in human and yeast cells found in recent years . The purpose of this study : to further clarify PinX1 the role and mechanism of regulation of telomerase / telomeres and telomerase-positive tumor cells . Method : mRNA sequences design PinX1 of shRNA under PinX1 , transferred HT1080 HepG2 and U2OS tumors cells in stable expression order to silence they endogenous PinX1 of expression ; then stable tumor cell lines These shRNA PinX1 the cell growth rate and cell were detected aging , DNA damage - induced apoptosis sensitivity and telomerase activity and telomere length , followed by Myc - POT1 co-immunoprecipitation ( IP ) . Results : PinX1 times in human tumor cell lines in the expression silent PinX1 result in telomerase-positive tumor cell aging, improve telomerase-positive tumor cells to DNA damage -induced apoptosis , caused by telomerase-positive tumor cells telomeres shortening and dysfunction, reduce the combined endogenous protein components of telomerase and POT1 telomere . Conclusion: This study reveals PinX1 there are also positive role in the regulation of telomerase / telomere ; The constitutive expression PinX1 help telomerase -mediated telomere maintenance and promotion of the growth of cancer cells .
