The Use of MALDI-TOF Peptide Mass Fingerprinting to Study Proteins from Cyanobacterium Microcystis Aeruginosa PCC7806

Abstract


Proteomics, especially peptide mass fingerprinting method , we identified Microcystis aeruginosa PCC7806 protein . In the study, we used 2 - DE and PMF combined with strategy . Microcystis aeruginosa PCC7806 cultured in normal BG-11 medium , combined with trichloroacetic acid / acetone precipitation method by sonication extract algae protein . The two-dimensional electrophoresis to isoelectric focusing using 18cm IPG strips (pH 4-7), SDS-PAGE as the second . The silver stained protein digging point , and then digested with trypsin . After trypsin digestion of the polypeptide fragment and the substrate solution blending, spotting the MALDI-TOF mass spectrometer (BRUKER, ReflexTM Ⅲ) is loaded for further analysis . Showing the peaks of each protein spots obtained after the analysis , to identify and eliminate the pollution peaks and cut peak . The next step is called the standard peak , we get a mass peak of 168 protein spots , these mass peaks submit to the the NCBI / MASCOT database to find matches protein . Theoretical parameters were compared through the protein chart experimental parameters (pI and molecular weight) and online database search , we realized the search , identification of these proteins . We have achieved the following results : 187 proteins were identified , and according to the source of these protein species and their relationship with our study species ( Microcystis aeruginosa PCC7806) are divided into three groups . 168 protein spots in the mass spectrometry results , since there are 18 points of the match proteins each contain 2 , so of the 187 protein . Of the 187 proteins identified , 14 (7.5%) protein derived from Microcystis aeruginosa PCC7806 ; 98 ( 52.4% ) of the proteins from different evolutionary tree belongs to the cyanobacteria Microcystis aeruginosa PCC7806 cyanobacteria belong branch ; 75 (40.1 %) of the protein , its classification with Microcystis aeruginosa our study is not closely related to , but consider other selection criteria , such as pI and molecular weight is selected . Compared with the latter two sets of data , the first group match the quantity of the protein is still low , indicating that other aspects of Microcystis aeruginosa .
