Abstract

Vibrio parahaemolyticus (Vibrio parahaemolyticus Vp) is a gram-negative halophilic bacteria, its cause of the disease, a high incidence of the popular range of serious harm. Vibrio disease is one of the most important factors of the limit of marine fish, shellfish and other marine economic development of the animal breeding industry, many reports of our fish, but few reports shellfish. Coelomactra invertebrates can not produce immunoglobulin, and the lack of specificity of antibody-mediated immune response. Studies have shown that an appropriate immune stimulant living organisms can increase non-specific immunity, and improve their overall disease resistance function. The outer membrane protein OMP (outer membrane protein) is one of the effective bacterial surface antigen, bacterial OMP vaccine prepared to protect the body against the attack of the same type or shaped bacteria having immunogenic and can induce a protective immune response has been confirmed, and the use of modern biotechnology easily extracted OMP, therefore developed development OMP vaccine has broad prospects. Monitoring of bacterial resistance and resistance mechanisms have become very active worldwide hot topic to study a variety of pathogens, antibiotic resistance and for resistance monitoring and a variety of bacterial disease prevention provide a scientific basis. The experimental by Vibrio parahaemolyticus Coelomactra, In view of this artificial infection continuous observation 7d, found the Vibrio parahaemolyticus Coelomactra highly pathogenic mortality increased with increasing infectious dose, the appearance of pathological changes significant take sick Coelomactra intestinal ax foot, outlet pipe, gonads, mantle do paraffin sections observed, each part there are varying degrees of pathological features, especially the epithelial cells change significantly; dodecyl sarcosyloxy the sodium extraction VP-OMP, SDS-PAGE method measuring protein molecular weight, the Bradford method determination OMP concentration, different culture conditions (temperature, pH, medium) have a different impact on the VP-OMP; incubation time of the VP-OMP No effect. Attack poison with VP-OMP immune Coelomactra is continuous observation of the 14d, measured the OMP on Coelomactra immune protection than inactivated whole bacteria seedlings, and high concentrations of OMP immune protection higher than the low concentration OMP immune protection rate, the immune indicators (antibacterial activity, lysozyme activity, phagocytic activity) has improved to varying degrees. Drug-sensitive paper Determination of Vibrio parahaemolyticus anguillarum mimicry Vibrio alginolyticus, Aeromonas hydrophila, Staphylococcus aureus, Escherichia coli seven kinds of bacterial resistance to 30 antibiotics found that different bacteria have different levels of resistance to different antibiotics; summary, this experiment pathogenic Vibrio parahaemolyticus Coelomactra, OMP (outer membrane protein) extraction and under different culture conditions SDS-PAGE, VP-OMP on non-specific immune Coelomactra preliminary study, and seven kinds of bacterial drug sensitivity tests for the precious shellfish pathological studies of bacterial diseases, OMP vaccine, shellfish and non-specific immunity and resistance of The monitor provides a theoretical basis.
