Purpose: the anti-malarial drug artemisinin is extracted from the plant Artemisia annua, is an efficient, quick, low toxicity, recognized by the World Health Organization is the only research and development of herbal medicine western medicine research standards. China is the world's largest producer of raw artemisinin drugs, its share of global production Jiucheng. At home and abroad mainly Artemisia annua artemisinin and its derivatives, and the study of the Artemisia annua other active ingredients and few reports. Artemisia annua with a variety of pharmacological activities reported in the literature on this topic in the protection of artemisinin extraction quality, combined with traditional medical records, to explore the other ingredients in the Artemisia annua, in order to achieve maximum utilization of Artemisia annua resources. This issue through procoagulant experiments in vitro, preliminary screening of the active site of Artemisia annua hemostasis objective experimental basis for the clinical application of hemostatic Artemisia annua. Meanwhile, the essential oil is also artemisinic except artemisinin outside a large class of substances, reported having significant antibacterial effect. Based on the above two points, our group proposed a new dosage form design: the hemostasis active site and Artemisia annua essential oils screened as the main material foundation, make it stop bleeding while both antibacterial effect. Depth study of the subject of Artemisia annua resources have some academic value and potential applications. Method: 1. The Artemisia annua vitro coagulation screening evaluation method: ① Artemisia annua extraction with 75% ethanol to give the crude extract. Crude extracts with petroleum ether, ethyl acetate, n-butanol sequentially extracted from a variety of organic solvent extract. A Comparative Study of the the coagulation plate method, tube method, plasma recalcification time assay in vitro coagulation experiments. ② screened ideal evaluation method, and is available with a laboratory hemostasis DRUGS This method was validated. 2. Artemisia annua hemostasis determination of the active site: (1) Artemisia annua crude extracts and four parts extract measured by coagulation plate method, tube method and plasma recalcification time. Of Artemisia annua butanol fraction were isolated and purified by polyamide, MCI, gel column. (2) plasma recalcification time as testing standards, n-butanol extract part too polyamide column eluted with water part again on MCI column fraction eluted with 20% methanol (denoted α) repeated in vitro coagulation experiments comparing research. (3) effective parts component analysis: the qualitative identification of the chemical, tannins, flavonoids, alkaloids, organic acids, amino acids, volatile oils, proteins, sugars, anthraquinones, saponins and other major categories of substances preliminary identification. 3 intends to develop novel antibacterial hemostasis formulations: First, part of the α and essential oils by mixing measured antibacterial and hemostatic activity; followed by design novel antibacterial hemostasis formulations, the draw more reasonable role model. Results: 1. The Chinese herbal coagulation screening evaluation model: Artemisia annua ethanol crude extract and four-part organic solvent extracts (petroleum ether, ethyl acetate fraction, n-butanol, water part) of part three in vitro coagulation experiments method comparison studies obtained Artemisia annua crude extracts in vitro coagulation effect, and n-butanol partial thromboplastin time highest rate. Plasma recalcification time Comparatively, with the clotting time assay is sensitive to the clotting time of plasma coagulation rapid characteristics, the experiment is conducted in a certain temperature, is continuously rather than intermittently observed more clearly, the end of the experiment, the results can be replication. 2. Of Artemisia annua hemostasis active site to determine: repeated comparative study of the in vitro coagulation experiments prove Artemisia annua butanol extraction part and α part has good in vitro procoagulant role. ② the active site in the screening process, the rate of clotting time is as follows: Artemisia annua crude (8.51%), the Artemisia annua butanol fraction (14.89%), n-butanol part too polyamide column fraction eluted with water (denoted as green is water: 22.11%) the the α part (27.37%), Sc, Sd, and the remaining partial thromboplastin time rate significantly lower than the the Artemisia annua crude extract even a non-condensable. α portion shorten the clotting time was significantly greater than the pre-extracted artemisinic bold was also greater than the subsequent extraction section. Therefore, the effective part of α part of Artemisia annua hemostasis. ③ qualitative identification results show that: the initially identified the main component of the effective parts of tannins, alkaloids, organic acids, sugars, anthraquinone several categories. New antibacterial hemostatic patch study: ① the alpha part of the essential oils mixed 1:1, determination of its antibacterial and hemostatic activity. The results show that the combination of the two respective effect is not weakened, on the contrary a certain degree of enhancement. (2) optimize the the hemostasis model of the proposed development of novel antibacterial derived theoretically better four patch model. By being liner, the fraction α-containing pressure sensitive adhesive layer, the essential oil layer and the release layer. Conclusion: the Artemisia annua crude ethanol extract through a series of extraction over polyamide column, MCI gel column, thin-layer chromatography separation and purification means filter finalized α part of Artemisia annua hemostasis active site; through essential oils and α part mixed preliminary experiments verify the theoretical feasibility of both common as the material foundation to develop formulations with antimicrobial hemostatic properties. The same time, the design of the four-patch model provides a clear direction for this new antibacterial hemostatic patch development. Artemisinin is mainly present in the the artemisinic portion of the petroleum ether extract, and the essential oils used in the present subject can be obtained from the pretreatment of the extract artemisinin, α part can be separated in the residue after the extraction of artemisinin. Therefore, the research of this subject, on the one hand while on the body for hemostasis to prevent infection, wound hemostasis to find a convenient and easy the The more ideal medicine antibacterial hemostatic dressings foundation; other hand, since the active material can expand as extract artemisinin byproduct, to a certain extent on the comprehensive utilization of Artemisia annua.
