Abstract


Newcastle disease (Newcastle Disease, ND) is an acute, contagious disease by the paramyxovirus family, a variety of birds caused by Newcastle disease virus in poultry mumps virus genus (Newcastle Disease Virus, NDV) infection and death . NDV has caused in the world within four popular supplies poultry industry has caused huge economic losses. 20 NDV isolated from Jiangsu Province envelope glycoprotein (F, HN) gene sequencing and genetic evolution analysis, the study of the biological characteristics of some strains selected according to the analysis results. For molecular biology information rich genotype Ⅵ NDV, and then for an in-depth understanding of the pathogenic properties of the virus of genotype Ⅵ basis of this study were separated into four pigeon genotype Ⅵ virus Determination of the complete genome sequence and analysis, and host specificity and virulence of the possible causes of the differences of pigeon genotype Ⅵ virus based on these results discussed. 1. Thus the currently Ⅶ d subtypes flocks in China is still very popular, and the vaccine strains infected flocks, NDV popular in China have shown a variety of genotypes coexist situation. The entire coding region of the HN gene amplification, cloning, sequencing final 20 NDV HN gene coding sequence. Statistical analysis revealed that the length of their encoded polypeptide gene? Isolates against NDV envelope glycoprotein (F, HN) monoclonal antibody emission spectrum experiment results show: HN protein monoclonal antibody (6B1) do not react with strains growing trend. 2. Si strains of pigeon Newcastle disease virus whole genome sequence of the determination and sequence analysis of RT-PCR technique the four pigeon NDV the whole genome sequence carried the amplification, by cloning, sequencing to obtain the four virus the whole genome sequence . The sequence analysis showed that the four virus full-length are 15,192 nucleotides (nucleotide, nt), 6 insertion position with the same genotype Ⅶ virus. Phylogenetic tree based on genetic drawn by the F gene hypervariable region 4 virus with Europe and Japan between 1987 and 2002 isolates belong to the same EU / re (recent European, EU / re) branch Ⅵ b subtypes shows The correlation of these regions in the epidemiological aspects. The full genome sequence of the F gene and HN gene were drawn three genetic phylogenetic tree highly consistent, indicating that the genetic evolution of the NDV genome-wide level. Genotype Ⅵ similarity between the protein amino acid sequence analysis of the other genotypes virus: variation of the P gene of NDV genes, L gene is relatively conservative; of the genotype Ⅵ virus virus type II gene similarity lower ; higher similarity with the gene Ⅳ, Ⅶ, Ⅷ virus, which reflects the genotype Ⅵ virus gene Ⅳ, Ⅶ VIII virus exists a certain correlation between the genetic evolution. Comparative analysis on the the pigeon virus from chicken virus gene coding region of the amino acid sequence found pigeon strains on the NP, P, L, there are several characteristic amino acid sites are NP protein 463 (N → D), 469 (P → I) and 477 (P → H) bit; proteins P-10 (D → E), 93 (TK / R) and 163 (R → G) bit; HN protein 122 (G → R) bit; L protein of 1110 (L → T / A) and 1757 (A → V) bit. M, F protein did not find the specific amino acid sites, found only in the HN protein (G122R). The pigeon strain specific amino acids appear likely to change the structure and biological characteristics of the virus-related protein, and thus impact its virulence and host specificity, related validation studies ongoing.
