Abstract


Populus (Populus euphratica Oliv) have a strong salt tolerance , preliminary studies show Populus xyloglucan transglucosidase gene expression significantly increased under salt stress , suggesting that this gene may have some kind of contribution to the salt tolerance of Populus . Populus for materials the the published GenBank database , Populus carried tremula x Populustremuloides , Solanum lycopersicum , Fagus sylvatica species such as xyloglucan transglycosylation the enzyme nucleotide sequence primers were designed to open its cDNA obtained by RT-PCR, code reading frame ( ORF ) . By sequence analysis shows that the ORF of length 870 bp , encoding a isoelectric point of 5.16 to 32166 Da molecular weight protein composed of 289 amino acids . Populus xyloglucan glucosidase gene (PeXET) built into the plant expression vector under the control of the constitutive promoter pGreen-0029 , transformed tobacco results that the turn PeXET tobacco salt tolerance and promote root elongation long role.
