Abstract

Corn stalk rot in corn production areas in China and the world has generally occurred. In recent years, changes in farming systems (such as conservation tillage, straw, etc.) and the effects of climate change, our corn stalk rot occurrence is gradually increased. A branch of the main pathogens of corn stalk rot Fusarium [of Fusarium verticillioides (Sacc.) Nirenberg] not only infect the root, stem and ear of corn, causing seedling blight, stalk rot and ear rot also as endophytes as both colonize the plant body without causing illness. In this study, using Agrobacterium tumefaciens in the red fluorescent protein gene the DsRed transferred to Verticillium strains of Fusarium FV through physiological and biochemical transformants screened transformants with wild mushroom smallest difference for subsequent analysis. The main conclusions are as follows: Agrobacterium-mediated transformation to random insertion of the GFP and DsRed fluorescent protein marker gene within the genome of Fusarium, Verticillium and exogenous gene differences are mostly random insertion of a single copy. Determination of physiological and biochemical indicators of transformants found DsRed more likely to lead to the emergence of mutants. Agrobacterium-mediated transformation into Verticillium strains of Fusarium FV DsRed red fluorescent protein gene by fluorescence microscopy observation the Verticillium Fusarium inductance corn inbred lines Root Colonization and growth law. Susceptible materials in the soil Verticillium the Fusarium first started from the fibrous, they may be a large number of fibrous roots colonization without causing the fibrous lethal. Root in the the Verticillium Fusarium infection corn maize resistant infection may play a key role, especially in the early stages of infestation. Followed by the Verticillium Fusarium along the taproot up the infection, and extended to the ground mycelial form of organization. Attached to the texture of the root epidermis, some spores germinate to form mycelium and expansion; others inward infected adherent cells, and then continue to infect the surrounding. Up by the root internal infection hyphae more upstream along the intercellular space, some will travel between different cell. Verticillium Fusarium infection susceptible corn sometimes form a mosaic structure, and this mosaic structure in the late mycelium fills the entire plant cells, resulting in cell death. The mycelium usually colonize fibrous roots below or near the cell. Resistant maize, the number of its fibrous roots was significantly lower than the susceptible materials, and the formation of necrotic spots to limit the expansion of Verticillium Fusarium. Analysis inoculated with different time Verticillium and Fusarium found in the number of colonies (CFU) of corn root and stem tissue formation, Verticillium Fusarium in the roots of the CFU values ??gradually decrease over time, while the the stem base CFU value was gradually increased trend. This shows Verticillium and Fusarium in soil through the root system infection pathway endanger aboveground organizations. Roots and stem base CFU values ??are lower than the susceptible materials resistant materials.
