Cryo-preservation has recently become a focal point of biological sciences and computational modeling, as the needs to protect and preserve various transplantable organ and cells has seen a steady increase. From stem cells, oocytes to skin and kidney, the effective storage of these cells, tissue or organ is decisive to the medical endeavors like tissue engineering and artificial insemination.In recent years, both Numerical modeling and experiment method has been introduced to this endeavor in the hope of discover optimal protocol of cooling procedure so as to augment cell surviving rate. As a quantitative branch of this endeavor, numerical simulation of species transportation during the cryo-preservation process could serve at least three purposes: Serve as an inexpensive replacement for vivo experiments; Help optimize the cyro-preservation protocols. Help to confirm or disprove intracellular ice formation theories.Since 1980s, the development of computational method has seen an explosive development in computational fluid mechanic, dynamic meshing, multi-element diffusion and porous simulation method. Endorsed by these progresses in the compotation field, a novel 2D Axis Symmetric model is developed here to address three major unsettle issues in cyro-preservation simulation. Firstly, examining the rapid dehydration induced gradient ring, which may in turn affect the intracellular ice formation probability. Secondly, investigate the influence of nucleus that may impose on cytoplasm transportation during cooling process. Finally, reflects the uneven transportation in heterogeneous cytoplasm and nucleoplasm.Famed Computational Fluid Dynamic software Fluent and Visual C was used to build this model. The species transport module of Fluent was coupled by Intracellular Ice Formation module, Moving Boundary module, Diffusion and Viscous module in VC to cooperatively run the simulation. Spatial graph of cyroprotective agent (CPA), water, ions distribution is achieved. The model itself is also one of most complete and complex model in cell cyro-preservation simulation, since it would likely to be the first 2D Axis Symmetric moving boundary model with nucleus of such kind.
