Background and purpose: breast cancer (breast cancer) is a common female malignancy, in recent years, with the rapid rise in its incidence, breast cancer has become an important disease threat to the majority of women's health. Recent studies suggest that the tumor is a stem cell disease, the abnormal tissue formed by the development of tumor cell proliferation with a tumorigenic ability. Tumor cells, only a very small number of cells with a tumorigenic potential, their numbers, though small, but can be unlimited proliferation, and may be symmetrically split self-profile or an asymmetric division generates ordinary tumor cells with a tumorigenic ability and therefore have stem cell characteristics , called cancer stem cells. Stem cell research to promote the development of the pathology and oncology. People began to realize that the tumor is a stem cell disease. Breast tumor disease, studies have proved the presence of mammary epithelial stem cells. Cancer stem cells in tumorigenesis and progression played a decisive role; no or only limited proliferative capacity and other tumor cell death by transient differentiation. The formation and development of breast cancer is a multi-step, multi-gene combined result of. Tumorigenesis learn that, an important link in the process of tumorigenesis is the occurrence of various types of gene expression changes that intracellular proto-oncogene activation and overexpression and inactivation of tumor suppressor genes. Therefore, we investigate the apoptosis inhibition gene Survivin, PTEN and embryonic stem cell gene Oct4 expression in human breast cancer and related breast cancer malignant research from the molecular level, the process of breast cancer stem cells and oncogenes, suppression changes in oncogene expression, reveal deeper lesions of the breast development and the evolution of new ideas. Methods: SP immunohistochemical method to detect different levels of the 37 cases of breast cancer, Survivin, PTEN and Oct4 expression and to analyze the correlation explore Oct4 expression in breast cancer tissue and its relationship with differentiation . Did not receive all the specimens preoperative radiotherapy and chemotherapy. Results: The positive expression of Survivin protein in breast cancer rate of 70.3% (26/37), the expression of survivin and the size of the patient's age, tumor size and ER, PR level axillary lymph node metastasis was no significant correlation (P gt ; 0.05). 2, PTEN protein in breast cancer tissue positive expression rate was 48.6% (18/37); With an increased breast cancer histological grade, PTEN protein expression rate downward trend, but not statistically significant. PTEN protein expression levels in breast cancer ER level was positively correlated (P lt; 0.05), with axillary lymph node metastases and the size of the patient's age was negatively correlated (P lt; 0.05), with no significant correlation between tumor size and PR levels. 3, Oct4 expression in breast cancer tissue is closely related to the degree of differentiation of breast cancer, breast Oct4 positive expression rate was 43.2%, poorly differentiated breast cancer Oct4 positive expression rate was 83.3%, significantly higher than the well-differentiated breast cancer ( 25.0%), a statistically significant difference (P lt; 0.05). 4, Fisher's exact test analysis drawn: between PTEN and Survivin expression in breast cancer as a negative correlation (exact test P = 0.013 lt; 0.05); of Oct4 with Survivin expression in breast cancer was significantly related (exact P = 0.01 lt; 0. 05). Oct4 and PTEN expression in breast cancer is no significant correlation (exact test, P = 1.00 gt; 0.05). Conclusion: Survivin and Oct4 expression in breast cancer was significantly related. Survivin expression in the expression of Survivin negative the Oct4 positive expression group and Oct4 negative expression group there are significant differences. Both PTEN and Oct4 was no significant correlation between PTEN and Survivin was a negative correlation of Survivin overexpression is associated with low expression of PTEN may be prompted Survivin over-expression in breast cancer stem cells from normal to cancerous process can promote PTEN mutation or deletion of PTEN arrest cell cycle, in turn weakens the induction of apoptosis, thereby preventing the disappearance of Oct4 retain abnormal self-renewal capacity, its unlimited amplification. 2, the level of expression of PTEN in breast cancer ER level was positively correlated with axillary lymph node metastases and the size of the patient's age was negatively correlated. PTEN and Survivin expression in breast cancer showed that the expression of PTEN and Survivin genes with negative correlation, both jointly involved in the occurrence and development of breast cancer, joint detection can be used as a reference to determine the degree of malignancy and prognosis indicators . 3, Oct4 expression in breast cancer tissue is closely related to the degree of differentiation, suggesting that Oct4 degree of malignancy of breast cancer, may be one of the important biological indicators to assess the prognosis of breast cancer.
