Exploring of Time Effect of SU11274 Impacting on Biological Behavior of Esophageal Carcinoma Eca109 Cells

Abstract


Objective: This paper aims to study the impact of the c-Met inhibitor SU11274 biological behavior of cell lines of Eca109 , and its target of c-Met protein and C-met mRNA circadian clock gene Clock mRNA could potentially contact , further explore the hour effect in its role in the process . Methods : (1) in vitro the culture of Eca109 cell lines , the expression of c-Met protein using immunocytochemistry . (2) with SU11274 (IC50 20nM) handle Eca109 cells , the by MTT growth curve , scratch experiments , transwell invasion chamber and the FCM, observed drugs on cell growth, migration , the invasion abilities as well as the impact of the rate of apoptosis and cell cycle . ( 3 ) in vitro the culture of Eca109 cell strains were detected at different time points Clock with C-met mRNA expression by RT-PCR method . ( 4 ) each time point c-Met protein expression detected by flow cytometry . ( 5) different time points SU11274 treated cells after 24 h detected by flow cytometry cell cycle . Results : (1 ) Eca109 cells expressing c-Met protein . (2) SU11274 experimental group, cell growth, migration and invasion capacity than the control group , the apoptosis rate was higher to the proliferation index lower than that of the control group . ( 3 ) within 48h Clock gene expression was rhythmic fluctuations ( P lt; 0.05 ) . (4) 24h within the Clock with C-met gene expression was similar rhythmic movements ( P lt; 0.01 ) . ( 5 ) within 24h c-Met protein expression was rhythmic movements (P lt; 0.05), a peak expression time point and C-met mRNA expression peak time point consistent . (6) at different time points SU11274 treated cells, the inhibition rate of cell proliferation index (P lt; 0.01). Conclusions : (1 ) c-Met inhibitor SU11274 can inhibit the growth and proliferation of Eca109 cells weakened the ability of migration and invasion , and increased apoptosis . ( 2 ) confirmed the expression of the in vitro culture the cells of Eca109 Clock change over time there are rhythmic . ( 3) The first of Eca109 cell C-met mRNA and c-Met protein expression was rhythmic fluctuations within 24 hours . (4 ) at the different time points by SU11274 treatment , cell proliferation inhibition rate index , Tip Tip hour esophageal cancer targeted therapy to consider effect .
