Objective To research the relationship of H.pylori infection and the expression of the Foxp3、TGF-β1 and IL-10 in human gastral mucosa.To provide the experiment evidence for H.pylori immune escape by inducing the expression of the Foxp3、TGF-β1 and IL-10 to inhibit immune.Methods1. We collect the selected 110 patients’gastric mucosa by biopsy forceps sterilized.All samples were put into non-gastric cancer group(80) and gastric cancer group(30) ,according to the results of the gastroscope and biopsy,which were divided into H.pylori positive group and H.pylori negtive group furtherly,in terms of the results of the H.pylori extaction,cultrue, or the specific PCR and urease test.2. The hybridization in situ was used to determine the expression of Foxp3 mRNA and TGF-β1mRNA in GA mucosa.The ELISA was applied to find the expression of IL-10 in GB mucosa.3. We prepared the model of the gastral epithelial cell strain infected by H.pylori in vitro.The ELISA was adopted to investigete the expression of TGF-β1 and IL-10 in culture solution.Results1. The H.pylori infection rate is 76.3% in non-gastric cancer group,and the rate is 63.3% in gastric cancer group.The total infection rate is 72.7%.2. In non-gastrial cancer group 44 samples are positive for the expression of the Foxp3 mRNA and 36 samples are negative.Comparision to the gastrial cancer group, there are 22 Foxp3 mRNA positive samples and 8 negative.The difference have no statistical significance(p>0.05). In H.pylori infection group,the Foxp3 positive rate is 84.2%,and the rate is 43.3% in non-H.pylori infection group.The significant difference is found by statistical analysis(p<0.05).3. In non-gastrial cancer group 51 samples are positive for the expression of the TGF-β1 and 29 samples are negative. Comparision to the gastrial cancer group, there are 22 TGF-β1 positive samples and 8 negative.The difference have no statistical significance(p>0.05). In H.pylori infection group,the TGF-β1 positive rate is 75.0%,and the rate is 43.3% in non-H.pylori infection group.The significant difference is found by statistical analysis(p<0.05).With regards to Foxp3 and TGF-β1 ,the coefficient correlation is 0.872,which has the statistical significance(p<0.01).4. In H.pylori infection group,the expression of the IL-10 is(3.03±0.88)pg/ml;In non-H.pylori infection group, it is(0.82±0.26)pg/ml; The expression of the IL-10 in H.pylori infection group is much higher than non-H.pylori infection group(p<0.01).5. The secretion of the TGF-β1 increased when GES-1 and H.pylori were co-cultured promptly. The expression of the TGF-β1 climbed the peak at 2 hours,then kept high concentration until 12 hours. The expression of the IL-10 increased at 1 hour gradually,which reached the climax at 1.5 hours,and then declinded fastly.It decreased by the original level of the co-culure,which was kept until 12 hours.Summary1.The H.pylori infection rate is about 72.7% in Shanxi,which is higher than the average H.pylori infection rate in China.2. The expression of the Foxp3 in H.pylori infection group is higher than that in non-H.pylori infection group,which indicate lots of Foxp3+Treg infiltrated in gastral mucosa infected by H.pylori maybe participate in the T cells proliferation and the function inhibition.3. The expression of the TGF-β1 mRNA and IL-10 protein in H.pylori infection group are higher than that in non-H.pylori infection group. In vitro,the expression of the TGF-β1 and IL-10 in the H.pylori infection group are higher than that in control.The identification of the results of the research in vitro and in vivo suggest the H.pylori infection may induce the secretion of the TGF-β1 and IL-10.4. The expression of the Foxp3 mRNA may be correlated with the TGF-β1mRNA. Conclusion Our research reveal the H.pylori infection maybe direct or indirect to induce the expression of the TGF-β1, IL-10 and Foxp3+Treg increasing, which can inhibit the immunity in gastral mucosa. Above all, it provide the microenviroment for the H.pylori infection sustained.
