Objective: This paper aims to study in rats ultrasound-guided percutaneous ethanol injection of liver surgery (PEI), the recovery of liver function and liver tissue regeneration and regeneration characteristics of existence. Methods: 40 SD rats, preoperative intraperitoneal injection of 1% pentobarbital 35mg/kg, anesthesia, using IU22 type ultrasonic diagnostic apparatus, the probe frequency 5MHZ-12MHZ for liver-guided, using a 1ml syringe, the 4th Semi-needle will slowly injected rat liver ethanol middle of each dose of 0.1ml. After injection, respectively, in the first days of 1,3,5,7 blood, serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) levels, in order to observe the recovery of liver function; take complete blood immediately remove the liver, surrounding the injection site was cut normal liver tissue, liver tissue and the liver capsule strip, into 0.01M phosphate buffered saline (PBS) were washed, cut into pieces, collagenase digestion, made into single cell suspension with 80 % ethanol. Washed twice with PBS after fixation, iodized propidium (PI) and 0.1% RNA enzyme (RnaseA), the line flow cytometry percentage of liver cells in S phase (S phase of liver cells / total number of viable cells); ethanol injection parts of the surrounding liver tissue in 10% formalin fixation, dehydration, paraffin sections, HE staining of liver tissue was measured mitotic index (MI), Feulgen staining DNA content was measured as well as immunohistochemical detection of proliferating cell nuclear antigen in liver tissue (PCNA). Results: PEI postoperative days 1 and 3 in rat liver mitotic index, DNA content, PCNA-positive cells and the amount of cells in S phase ratio was significantly higher than the preoperative (P lt; 0.01), and reached its peak in the first three days: MI of (1.76 ± 0.20)%, SPR was (16.53 ± 2.96)%, DNA content of (8.68 ± 0.94)%, PCNA was (11.08 ± 1.65)%; first 5 days of the reproduction parameters were significantly reduced, on the 7th day restore the preoperative level. Rat liver function after surgery increased significantly at day 1, AST was (417.02 ± 25.07) U / L, ALT was (357.93 ± 26.83) U / L, then decreased until the seventh day to restore the preoperative level . Conclusion: PEI rat liver after injection with ethanol dehydration, coagulation, and cause blood clots, can directly cause dehydration rat liver tissue coagulation and necrosis, postoperative ethanol diffusion limitations, may also cause distant liver cell necrosis, leading to impaired liver function, and further promote its surrounding liver tissue regeneration to some extent. The regeneration and liver function recovery law rules on clinical application of percutaneous ethanol injection in the treatment of intrahepatic liver cancer have a certain significance. Ultrasound-guided percutaneous ethanol injection sclerotherapy of the liver with less damage, good repeatability, efficacy, side effects, simple operation, quick recovery, easily accepted by patients, etc., so it has broad application prospects.
