Abstract


Objective To explore the to wistar rats asthma airway remodeling model of glucocorticoid matrix metalloproteinase-9 , endothelin-1 and its use of glucocorticoids on improve asthma wistar rat airway remodeling clinical value . Methods 40 wistar rats were randomly divided into a control group (C group ) , model group (N ) , the early intervention group (E ) , the late intervention group (L ) , n = 10 . Ovalbumin sensitization and inhalation challenge rat asthma model . Model group on days 1,8 injection of ovalbumin ( 100 g / L ) , 15 days before atomization ovalbumin ( 10 g / L ) , 20min / d ; early intervention group with model group , but 1h before atomization ovalbumin intraperitoneal injection of dexamethasone ; late intervention group to aerosolized ovalbumin 2 weeks after injection of dexamethasone ; atomization and injection with saline instead of the control group . Eight weeks of continuous atomization . Within 24 hours after the last challenge anesthesia thoracotomy . Immediately right heart blood , rat plasma endothelin-1 content was measured using a specific radioimmunoassay . Right lung and trachea fixed , embedded preparation pathological . Lung tissue stained by hematoxylin - eosin staining ( HE ) , Masson staining and matrix metalloproteinase -9 (MMP-9) by immunohistochemical staining . Computer color image analysis to measure the morphological parameters of rat airway . Results of the early intervention group smooth muscle thickness , the Explorer wall thickness and collagen area were higher, and were significantly lower than the model group and the late intervention group ( p lt; 0.01 ) . Late intervention group smooth muscle thickness , model group ( p lt; 0.01 ) the Explorer wall thickness and collagen area below . Early intervention group, lung tissue matrix metalloproteinase -9 expression and plasma endothelin-1 level higher than that of the control group and lower than the model group and the late intervention group ( p lt; 0.05 ) , the late intervention group lung tissue matrix metalloproteinase-9 expression and plasma endothelin -1 levels lower than the model group ( p lt; 0.05 ) . Rat airway remodeling conclusion asthma model , glucocorticoids affect matrix metalloproteinase-9 , endothelin-1 expression and reduce the deposition of collagen in the extracellular matrix deposition alleviate , inhibit the airway wall , smooth muscle thickness increase and impact airway remodeling . Pathogenic environment persists , glucocorticoids can only delay the process of airway remodeling , can not be reversed .
