The Intestigation on Diagnostic Value of BALF-phage-based Splitting Assay in Fast Detection for Mycobacterium Tuberculosis

Abstract


Objective: To investigate lavage fluid (BALF) phage lysis method (PhaB) Rapid detection of Mycobacterium tuberculosis clinical value . Methods: 30 cases of pulmonary tuberculosis patients and 30 non- TB patients ( including lung cancer, 12 cases , 5 cases of pneumonia , chronic obstructive pulmonary disease, 13 cases ) lavage specimens phage lysis experiments with other TB screening method ( sputum smear acid-fast staining , Roche sputum culture , smear acid-fast staining lavage fluid , bronchoalveolar lavage fluid Roche culture , PPD, TB-AB) controls. Results: 30 cases of pulmonary tuberculosis phage lysis lavage fluid positive in 18 and negative in 12 cases ; lavage fluid acid-fast staining positive in 9 cases, negative in 21 cases ; lavage fluid culture positive for M. tuberculosis in 20 cases, negative 10 cases ; sputum smear positive for acid-fast staining in 6 cases, negative 24 cases ; sputum culture positive in 11 cases, negative in 19 cases . 30 cases of non- pulmonary tuberculosis and tuberculosis antibody PPD test (TB-AB) false- positive cases were detected in 10 cases and 7 cases , and phage lysis lavage fluid , bronchoalveolar lavage fluid smear acid-fast staining lavage Roche culture fluid , sputum acid-fast staining and culture false positive cases were 0 cases ; lavage phage lysis sensitivity 60.0% , and acid-fast staining of sputum , sputum culture Roche , lavage smears acid-fast staining compared , the difference was statistically significant ( three caught P lt; 0.05); Roche and bronchoalveolar lavage fluid culture wears the difference was not statistically significant (P gt; 0.05); its specificity was 100 % higher than PPD test and tuberculosis antibody detection ; this method is 100% positive predictive value , negative predictive value of 96.8% . Conclusion: BALF phage lysis method detection of Mycobacterium tuberculosis fast, simple and can be used as an important method of diagnosis of tuberculosis .
