Abstract


Ganoderma lucidum fungus is the dry fruit body of Ganoderma lucidum (Ley-ss.ex Fr) Karst. or Ganoderma sinense Zhao, Xu et Zhang of Polyporaceae . A large number of pharmacological studies have shown that Ganoderma lucidum has the activity of immune regulation, anti-tumor, anti-aging and improving the hypoxia activity of the body. But it leads to larger differences in the quality of the crude drug because of the ecological environment, species, collection time, processing technique are different. So it is necessary to study the method of controlling the quality of Ganoderma lucidum and provide experimental foundation for controlling the quality of Ganoderma lucidum in comprehensiveness and systematism.Due to the complicated geographical and climatic conditions, the case of multi-origin is so widespread in Ganoderma lucidum’s species that their species are complicated for a long time. Ganoderma lucidum is mainly bought from the Chinese crude drug market and the Chinese herbal pharmacy for the domestic consumers, and so the Chinese herbal samples we used in the experiment were mainly from these two paths. The results provide experimental foundation for the identification and quality control of Ganoderma lucidum.In this paper, the contents of this experiment were divided into three parts, including quality standards, The high-performance liquid chromatographic (HPLC) fingerprints and infrared fingerprint:1.Study on quality standards: The physical and chemical testings of Ganoderma lucidum from different paths were performed, including the determination of moisture, total ash, acid insoluble ash, extractum and polysaccharide content of Ganoderma lucidum. And these samples were studied and evaluated comprehensively from both qualitative and quantitative aspects.2.Study on the fingerprints: HPLC fingerprints analysis method for the triterpenes of Ganoderma lucidum was set up preliminarily. The best chromatographic conditions were optimized including the column, the mobile phase systems, detection wavelength, column temperature, etc. The column was Dikma Diamond-C18 (4.6 mm×200mm,5μm) ; gradient elution with a mixture of two solvents: solvent A, 0.1% acetic acid and solvent B, acetonitrile; the flow rate was 0.8ml·min-1; the detection wavelength was 254nm and the column temperature was 35℃.The HPLC fingerprint of Ganoderma lucidum was established preliminarily with good specificity, characteristic, stability and reproducibility.3. Study on infrared spectra: Ganoderma lucidum was studied through infrared spectrometer and the cluster analysis method was used in the data analysis, it provides the experimental foundation for the identification.Through this research, we hope to provide a workable standard for the quality control of Ganoderma lucidum and regulate the circulation of Ganoderma lucidum from the Chinese crude drug market. And we also hope to provide a reference foundation for the 2010 edition of Pharmacopoeia which will have the characteristics of standardized, systematic, integrity, pragmatic.
