Objective:To assess he efficacy and safety of Misoprostol while intrauterine insemination. Method:The Cochrane Library (CENTRAL, No.3,2009), EMbase (1974-2009.10)、MEDLINE (1966-2009.10)、SCIENCE Direct Database(1966-2009.10)、CBM (1978-2009.10), CNKI (1989-2009.10), VIP (1989-2009.10) were searched for randomized controlled trials or clinical controlled trials. Statistical analysis was performed by RevMan5.0 software to do Meta-analysis. Result:Five Randomized controlled trials (RCTs) were included according to the inclusion and exclusion criteria at last and they are all published in English. Five RCTs involving 546 participants and 1133 cycles. Meta-analysis showed that 200 ug Misoprostol couldn’t increase pregnancy rate [RR=1.19 (0.60,2.36)] of IUI in groups of 200 ug Misoprostol vs placebo,400 ug Misoprostol could increase pregnancy rate [RR=1.95 (1.19,3.19)] of IUI in groups of 400 ug Misoprostol vs placebo and 400 ug Misoprostol also has higher abdominal cramp rate [RR=3.13 (1.86,5.26)] and vaginal bleeding rate[RR= 5.25 (2.08, 13.21)] in 400 ug Misoprostol vs 200 ug Misoprostol. Conclusion:Because of the studies about this area was too little, although there are evidence for Misoprostol could increase pregnancy rate, the incidence of adverse reaction was higher, it’s not recommend the use of Misoprostol in IUI cycles. Further high-quality, large-scale randomized controlled trials are still required.
