Abstract


Background: The tumor is the most common human a polygenic multifactorial disease the global cancer morbidity and mortality has been high, and showing a rising trend. The high rate of tumor invasion and metastasis is a key factor in patients with malignant tumors with poor prognosis and high mortality. Invasion and metastasis of biological processes involving a large number of tumor-related genes structural changes and abnormal expression of important biological characteristics of malignant tumors. Therefore, analysis of malignant tumor gene expression profiling and protein expression profile changes can understand tumor development and proliferation of related genes in tissues and found that the invasion of metastasis-related genes have important for improving the prognosis of patients with malignant significance. Recent years significantly Egfl7 enhanced cell motility migration stimulating factor is an important movement, and the protein is expressed in several tumor cell lines highly raised, suggesting that there may be a potentially important role in tumor invasion and metastasis. Has yet to see the Egfl7 in common solid tumors of epithelial cell tissue and cell lines as well as adult normal tissue expression profiling studies have reported, it is unclear whether Egfl7 involved in the invasion and metastasis of malignant. Objective: To detect Egfl7 in normal adult tissues, 10 common epithelial tumors and tumor cell lines corresponding mRNA and protein expression levels determine the expression profile; analysis of its cellular localization in tumor cells; explore its relationship with liver cancer and breast clinicopathological features and prognosis of cancer, preliminary to understand Egfl7 liver cancer and breast cancer recurrence and metastasis, screened to determine liver cancer and breast cancer potential prognostic markers for in-depth study in malignant Egfl7 role in the development lay the foundation. Methods: RT-PCR and Western blot detection of three cases of 19 adult normal tissues, 10 kinds of common epithelial tumor cell lines and 36 cases of liver cancer, 30 cases of breast cancer, 30 cases of lung cancer, 30 patients with colorectal cancer, 30 cases of gastric cancer , three cases of malignant gliomas, three cases of ovarian cancer, 6 cases of renal cell carcinoma, 4 cases of prostate cancer and 5 cases of esophageal cancer tissue specimens and corresponding adjacent tissue samples Egfl7mRNA and protein expression levels; cellular immunity The fluorescence analysis the cell lines Egfl7 protein distribution; using immunohistochemical method detected 132 cases of liver cancer, 102 cases of breast cancer and 30 cases of lung cancer, colorectal cancer, 30 cases of gastric cancer, 30 cases of malignant gliomas eight cases of ovarian cancer 6 cases, 16 cases of renal cell carcinoma, prostate cancer patients and 10 cases of esophageal cancer paraffin specimens Egfl7 protein levels and subcellular localization, compare Egfl7 mRNA and protein expression levels in common epithelial tumors cancer cancer and adjacent tissues difference, at the same time explore the relationship between liver cancer and breast cancer paraffin specimens protein Egfl7 and his clinical and pathological features. Combined with clinical follow-up analysis of the level of protein expression in Egfl7 prognosis of patients with hepatocellular carcinoma and breast cancer. Results: RT-PCR and Western blot results showed that Egfl7 mRNA and protein in epithelial tumor cells: mRNA and protein in normal adult tissues Egfl7 only a small amount of expression, the highest expression in lung, retinal vascular-rich organs and tissues; Department of higher levels of expression the; Egfl7 mRNA and protein expression levels in the epithelial tumor cancer tissue was significantly higher than adjacent tissues (P <0.05). Immunofluorescence and immunohistochemistry confirmed positioned Egfl7 protein in the cytoplasm of cancer cells. Epithelial tumors of cancer tissue Egfl7 protein expression were significantly higher than the adjacent tissues (P <0.05). Egfl7 expression with HCC and clinicopathological findings: Egfl7 with HCC nodules, envelopes, and without venous invasion significantly (P <0.05), with gender, age, liver cirrhosis, Edmondson-Steiner grade and tumor diameter size is independent of the clinical and pathological features; Egfl7 with breast cancer clinical TNM stage, lymph node metastasis, ER and Her-2 was significantly (P <0.05), but not with age, tumor size, pathological type, cell differentiation degree of and PR receptors unrelated to the clinical and pathological features. HCC and breast cancer clinical follow-up studies have shown Egfl7 intensity of protein expression was negatively correlated with liver cancer and breast cancer prognosis, the HCC patients high expression of Egfl7 postoperative overall survival and disease-free survival time were significantly shorter in the low expression group ( median survival time was 410 days of VS640 days, P = 0.001; vs560 days, the median survival time was 315 days, P = 0.007); Egfl7 high expression of breast cancer patients overall survival and disease-free survival time was significantly shorter than the low expression group (median survival time of 1900 days vs2710 days, P = 0.001; median survival time of 1450 days vs2110 days, P = 0.018). Multivariate Cox regression analysis showed that the Egfl7 protein expression is an important independent factor for liver cancer and breast cancer prognosis. Conclusion: for the first time established a 19 normal adult tissues, epithelial tumors and tumor cell lines Egfl7 expression profiling for the further study Egfl7 laid the foundation for the role of the tumor. For the first time analysis Egfl7 with HCC and breast cancer prognosis, the confirmed Egfl7 would affect an independent risk factor for liver cancer and breast cancer prognosis for the study provide an important basis Egfl7 as a new tumor marker substance monitoring tumor recurrence and metastasis, prognosis. First confirmed epithelial cancer cells, Egfl7 secretion, subcellular localization analysis in the cytoplasm, suggesting that Egfl7 is likely to be HCC, a new serum diagnostic markers can be used for the diagnosis of HCC, HCC metastasis and recurrence monitoring and prognosis judgment.
