The technology of Magnetic Resonance Spectroscopy (MRS) is a newly-developed means for analyzing some specific nuclei and their compounds based on the principles of magnetic resonance and the effects of chemical shift. It supplies us a method for measuring metabolite concentrations in the brain noninvasively and quantitively. and it can evaluate lesions at the molecular level . Currently, MRS can detect MR signal of 1H,31P,13C of human tissue, but only H-proton magnetic resonance spectroscopy (1H-MRS) is widely used in clinic. According to different sampling volume, 1H-MRS is divided into single voxel spectroscopy(SVT)and multi-voxel spectroscopy (MVT). SVT has been widely applied in quantitation of metabolites, but the application of MVT hasn’t been reported in China and the technical study about MVT is hardly seen home and abroad. It is noticed that the realization of the quantitation of metabolites using MVT and to the improving of the accuracy of quantitative techniques are of great significance for its application in clinic.This study includes several aspects. Firstly, we had studied the manul of quantitative software-LCModel to understand the meaning of the control parameters and its influence to the quantitation results, which have great significance to realize the quantitation using MVT. Secondly, the optimization of the control parameters were performed in phantom experiment. The relationship of the main parameters and the quantitation results was studied and accurate quantitation was achieved in phantom with known concentration by multi-voxel spectroscopy. Thirdly, the validation of this method was performed in clinical healthy volunteers and patients. And finally, the influence of total scan on the quantitaion was explored in SV scanning, providing the foundation for the precise quantitation in clinical.Accurate quantification with MV is a hot issue all over the world and is still in the initial stage. The paper on magnetic resonance spectrum absolute quantitation with multi-voxel spectroscopy techniques was not found in domestic research, and foreign related research was made by only a few group without encouraging progress. Absolute quantitation of MRS by MV is realized in this study with LCModel and SAGE software. The calibration factor was optimized and the influence of scan number to accurate quantitation was discussed. In phantom and clinical experiment, the absolute quantitation was made for diagnosing of diseases. We believe that the theoretical description of absolute quantitation and report about absolute quantitative technique in this thesis is helpful to the follow-up workers.
