Abstract


Purpose:In this experiment,we determined the contents and fingerprint of the target components in the different source and processed Cimicifugas which could help to study the Quality of the Cimicifugas , the study of pharmacokinetics and tissue distribution could discover the body process in rat.Materials and Methods: we have studied the best extraction by reflux and high performance liquid chromatography; made the qualitative identification of the five target components by TLC;determinated the contents of five target components by Gradient elution and Wavelength switching;compared the fingerprints of different source and processed Cimicifugas by HPLC;studied the pharmacokinetics and tissue distribution by HPLC.Results:1.We determined the best extraction was:20 times of 30% alcohol and reflux 1.5h.2. TLC:the five components was well separated.3. Determined the five components by HPLC,the results showed a good range of linear relationship, precision, stability and freeze-thaw frequency of visits are comply with the relevant requirements.4.We set up the fingerprints of Cimicifugas by HPLC,and evaluated the similary of different source Cimicifugas.5. Pharmacokinetics:the body process of Cimicifugas in rat comformed to Two-compartment model,and could find the two components in the organization of choice.Conclusions: In this experiment,we determined the best extraction;set up the methods of determining the five components at the same time, set up the Fingerprint;these studies can provide the basis for quality standards of Cimicifugas.We also studied the body process in rats of the unprocessed and processed Cimicifugas,and compared the differences and similarities by studying the Pharmacokinetics and Tissue distribution,which could help to provide the basis for Pharmacodynamics of Cimicifugas.
