Abstract

Objective: through the establishment of Chlamydia trachomatis F serotype reproductive tract infection of pregnant mice model, to understand the impact of the genital tract Chlamydia F-type infection on pregnancy outcome in mice. Understanding of the relationship between C. trachomatis infection in pregnant rats outcome of pregnancy and in vivo Th1-type, Th2 cytokine levels, and understand its immune status and pregnancy outcome, azithromycin intervention immune mechanism of adverse pregnancy outcomes Chlamydia trachomatis. Methods: All pregnant rats were randomly divided into three groups, in 2, 3 and 4 days pregnant control group A group vaginal inoculation Chlamydia infection of McCoy cells prepared SPG suspension 30ul, experimental group B group vagina inoculated with 1.0 × 10 6 IFU / ml Chlamydia trachomatis F-type strains SPG suspension 30ul, experimental group, Group C in eight days of pregnancy, the vaginal inoculation chlamydia after intraperitoneal injection of azithromycin 10mg/kg (single dose). Pregnant 12 days detected by ELISA in the serum of each group of mice Th1-type (IFN-γ, TNF-α) / Th2-type (IL-4, IL-10) cell factor expression levels of IL-4 and calculate / TNF -α, IL-10 / TNF-α ratio. By analysis of variance and χ2 test comparing three groups of pregnancy failure rate, embryo resorption rate, fetal weight, the amount of weight gain during pregnancy. Ratio and cytokine level and TH1/TH2 immune to adjust the balance between the three groups were compared. Results: Group B of the experimental group that F-type strains of mice pregnancy failure rate of the experimental group inoculated with Chlamydia trachomatis genital tract for 50% of group A was significantly high in the control group 13.3%. Repeated pregnancy failure rate of experimental group B group to 22% in the control group, Group A 0 compared, differences have significant; experimental group C group that azithromycin intervention group, the pregnancy failure rate and repeated pregnancy failure rate than that in group B low and compared with A group high, compared to the experimental group A decreased, but no significant difference (P gt; 0. 05) repeated pregnancy failure rate has dropped significantly, the significant difference in sex (P lt; 0, 05). 2 embryos absorption rate, embryo weight, the amount of weight gain during pregnancy compared to the three groups showed no significant resistance. 3 vaccination Chlamydia trachomatis group compared with the control group vivo Th2 cytokines the decline (IL-10), while the increase in Th1-type cytokines (TNF-α), IL-4 / TNF-α, IL-10 / TNF-α ratio experiment group was significantly higher than that of the control group, the formation of Th1-type immune bias. The azithromycin group of intervention to reduce the rate of pregnancy failure and reduced Th1 immune bias degree of. Conclusion: an experiment successfully cultured human non-gonococcal urethritis caused by Chlamydia trachomatis F serotypes, and amplification, sequencing and counting. 2 experiment is the first successful use of Chlamydia trachomatis F serotype infections genital tract of pregnant rats to establish an animal model to evaluate the impact of Chlamydia trachomatis on pregnancy outcome in pregnant rats, as well as studies of the immune status of pregnant mice to explore the pathogenic mechanism of Chlamydia trachomatis . Use the chlamydia F serotype infection of pregnant mice as an experimental method is feasible to establish an animal model. 3 Chlamydia trachomatis F serotype vaginal vaccination can lead to the pregnancy failure rate of pregnant rats and repeated pregnancy failure rate increased, while the of embryonic absorption rate, embryo weight, no significant effect on the amount of weight gain during pregnancy. The azithromycin intervention can reduce repeated pregnancy failure rate, and no significant improvement in the rate of pregnancy failure. 4 Construction of C. trachomatis F serotype vaccination of pregnant rats to induce the body to produce specific cellular and humoral immunity, cellular immunity is more significant, dominant Thl response embryo Taidu Th1-type cytokines, can interfere with early embryonic development cause pregnancy loss, leading to adverse pregnancy outcomes. Azithromycin intervention can reduce the degree of this bias, so as to improve the outcome of pregnancy. Revealed Chlamydia trachomatis pathogenic mechanism of adverse pregnancy outcomes.
