Abstract


Piglets Escherichia coli diarrheal disease caused serious economic losses to the world pig industry, has seriously hampered the healthy development of the pig industry. Enterotoxigenic Escherichia coli (enterotoxigenic Escherichia coli, ETEC) cause diarrhea the most important pathogen adhesin and enterotoxin caused by ETEC diarrhea is the most important virulence factors. Currently, many scholars at home and abroad of the disease pathogenesis, diagnosis and control a lot of research and made a lot of progress, which ETEC molecular biological characteristics of more. A lot of information that the prevalence of ETEC virulence factors with regional differences, so about an area popular adhesin and enterotoxin type Escherichia coli diarrhea in piglets in the region diagnosis, prevention and control of great significance. ETEC virulence factors, traditional methods of detection time-consuming, inefficient, low sensitivity, multiplex PCR detection ETEC virulence factors a fast, accurate testing techniques. Piglets gut microflora balance is very important for the health of the piglets, diarrhea and healthy intestinal flora of piglets microcosms comparative analysis of structural changes in, guiding significance for the prevention and treatment of the disease. In view of this, the study by multiplex PCR for the detection of molecular epidemiology, while taking advantage of the separation of the different regions in Shaanxi Province piglets with diarrhea stool samples of 104 strains of pathogenic E. coli adhesin and enterotoxin type ERIC-PCR technology to health molecular analysis, and diarrhea piglets intestinal flora structure in order to study the variation of the structure of intestinal flora in the diarrhea and the effects of the different types of toxins, has been tested and results are as follows: 1. multiplex PCR technology, separation identified 104 E. coli expression strains of enterotoxin 45, STA, STB, LT and STA number of the LT-positive strains were 28,5,8 and 4. Express adhesin of 31, K88, K99, 987P and K88 987P-positive strains were 4,22,2 and 3. In the sample is detected, while expression of the Escherichia coli enterotoxin and adhesin 14; southern Shaanxi region only detected Sta enterotoxin and K99 adhesin, northern Shaanxi region is more complex, of each toxin type; 1 to 7-day-old piglets with diarrhea Escherichia coli toxins expressed K99 and STA, 20 ~ 30 days to various toxins have different levels of detection. Healthy piglets and piglets diarrhea intestinal flora structure were detected and analyzed by ERIC-PCR technology, diarrhea piglets intestinal flora structure fingerprints have at least one advantage bands, indicating that there is a type of bacteria abnormal proliferation , resulting in the piglet gut microflora structural imbalance; carry different enterotoxin ETEC diarrhea sample analysis showed that these piglets intestinal flora structure very different expression the same enterotoxin samples microflora basic similar, suggesting that ETEC enterotoxin the piglet intestinal flora structure has a significant influence. These results suggest that, Shaanxi Province Spatial differences cause diarrhea epidemic strains of ETEC ETEC toxin type structure of intestinal flora significantly.
