The purpose of this experiment is to study overcast artery precursors of milk protein concentration of dairy goat mammary uptake and influence of the breast tissue estrogen receptor mRNA expression. Selection of 2-3 years of age, body weight, milk yield similar venous cannulation overcast external carotid artery and milk in dairy goats six, six sheep for six replicates, each sheep were perfusion amount of essential amino acids missing is formed, and on its own does not perfusion control. Using perfusion of arteriovenous concentration difference technique 3h (4.65g as 75% missing amount), 4h (6.2g 100% missing amount), 5H (7.75g of 125% missing amount) of the breast milk ingredient precursors intake , using molecular techniques to study milk protein precursor concentration changes in breast tissue estrogen receptor mRNA expression. (1) determination of the six dairy goat blood flow, and 6 sheep average breast blood flow 124.76L / d, and the average mammary plasma flow to 83.17L / d. The calculation of the ratio of mammary blood flow and milk yield, the results are basically in line with the theoretical value of 500:1. (2) overcast outside after intra-arterial infusion of essential amino acids increased breast intake of essential amino acids and non-essential amino acids. Perfusion of essential amino acids, the treatment group breast EAA intake was significantly (P lt; 0.05) higher than that of the control group, the difference between the treatment groups was not significant (P gt; 0.05), three treated Thr, Met, Lys intake amount are highly significant (P lt; 0.01) higher than that of the control group. Compared with the control group, the treatment group of Ser and Pro uptake differences not significant (P gt; 0.05), other non-essential amino acid intake treatment group were significantly (P lt; 0.05) higher than that of the control group. NEAA uptake of the treatment groups was significant (P lt; 0.01) higher than that of the control group. Treated glucose, triglycerides and total cholesterol significantly (P lt; 0.05) higher than that of the control group the urea nitrogen difference was not significant (P gt; 0.05). With the rise of the essential amino acid perfusion, milk protein and lactose content was significantly higher (P lt; 0.05), milk fat and non-fat solids content also has the tendency to increase, but the difference was not significant (P gt; 0.05). (3) the three treatment groups plasma IGF-I, GH, PRL significantly (P lt; 0.05) higher than that of the control group, treated in the best perfusion 100% missing the essential amino acids. Real-time PCR found that perfusion necessary after the end of the amino acids, the breast tissue IGF-IR, GHR, PRLR mRNA relative expression levels treated extremely significant (P lt; 0.01) higher than that of the control group.
