Abstract


Grape powdery mildew is a serious harm to fungal diseases of grape growing, breeding of disease-resistant varieties of resistance genes resources is one important way to solve the problem. Protein is the essence of life activities, the performance of the different resistance grapes is a decision by its specific proteome Proteome react more directly than the differences in the genome of the nature of plant disease resistance. Therefore, the use of proteomics comparative study of different disease-resistant grape proteome differences, to find a susceptible protein and disease resistance protein, then obtained directly related to the powdery mildew resistance gene, explore grape disease resistance mechanisms, the use of disease-resistant genetic resources in breeding disease-resistant varieties and disease problems solve grape production has important theoretical significance and practical value. In this study, this sense of powdery mildew of the Carignan grape varieties (paternal) and resistance to powdery mildew of grape varieties Shirakawa -35-1 (female parent) both hybrid obtained F1 generation of disease-resistant plants 6-12-4 and susceptible plants 6-12-2 as the material, 0,2,4 d sampling, and analysis of the different periods Proteome Resistance grapes inoculation treatment were inoculated with powdery mildew. The main results are as follows: (1) the initial establishment of a suitable two-dimensional electrophoresis system. Compares the different protein extraction method, two-dimensional electrophoresis of the different sample volume, different isoelectric focusing procedure, strips of different pH and different SDS-PAGE gel concentration effect. The results showed that for the use of pH 4 ~ 7, linear 7-cm IPG DryStrip 300μg of sample volume, the use of isoelectric focusing procedure B, and obtain a better two-dimensional electrophoresis using 12% SDS-PAGE gels are conducive results; while for strips of 17 cm on the sample volume is 1050μg. (2) through the two-dimensional electrophoresis technology to compare the protein material of the six different treatment group differences. Total of 26 differentially expressed protein spots, 10 of which express the recovery and purification of the protein spots were identified by mass spectrometry, seven positive spots, (1305,4708,4709,5411,5625,6915,7121 ), three negative spots (1222,1420,7615). (3) protein sequencing showed that the difference in spots, in obtained seven positive spots, the differences in the protein spots 1305 and 6915 is assumed; differences spots 4708 and 4709 unnamed protein; differences spots 5411 for the 33 KDa photosystem II oxygen composite vitro weeks protein; differences spots 5625 ribulose 1,5 - bisphosphate carboxylase / oxygenase; differences spots 7121 pathogenesis-related protein 10. These proteins may be related to the grape plants resistant and susceptible to powdery mildew certain characteristics.
