Abstract


Objective: Using RT-PCR and Western blotting technique (Western Blotting), detection of embryonic stem cell transcription factor Sox2 in lung carcinoma and adjacent lung tissue nucleic acid and protein levels are expressed and expression level was compared in lung tissue and cancer adjacent tissue level differences, analyze its relationship with pathological type, differentiation and clinical stage correlation investigate whether the development of lung cancer plays a role, and whether as a marker for early diagnosis of lung cancer. Methods: Surgical resection of lung cancer and adjacent lung tissue in 40 cases, including 20 cases of squamous cell carcinoma, adenocarcinoma in 15 cases, 3 cases of small cell carcinoma and large cell carcinoma in 2 cases; in - group of 23 cases of well-differentiated, poorly differentiated group of 17 patients; 15 cases of stage Ⅰ, Ⅱ of seven cases, Ⅲ of 18 cases. The cancer tissue and cancer tissue were used RT-PCR and Western Blotting detection techniques embryonic stem cell transcription factor Sox2 in nucleic acid and protein expression levels, the gel image processing system analysis of the target with the molecular weight and optical density values, the data were analyzed with t-test, analysis of variance and rank sum test, treatments with SPSS11.5 software package, based on the results of the final transcription factor Sox2 in lung cancer and adjacent tissues, and to explore its clinical relevance. Results: The four kinds of pathological types of lung carcinoma and adjacent lung tissue, embryonic stem cell transcription factor Sox2 in nucleic acid and protein expression levels consistent. In nucleic acid and protein levels were detected in embryonic stem cell transcription factor Sox2 expression, and lung tissues were significantly higher than in adjacent lung tissue. And in terms of histological type, degree of differentiation of view or from the clinical stage, Sox2 expression in lung tissue were higher than the adjacent lung tissue, and the difference was significant. Nucleic acid and protein in lung cancer clinical stage Ⅲ level of Sox2 expression levels were significantly higher clinical stage Ⅰ, Ⅱ period, and the poorly differentiated group of Sox2 expression levels were significantly higher than in the - well differentiated group, there were significant differences in the two groups. Two levels of different pathological types of Sox2 expression in lung cancer tissues showed no significant differences. Conclusion: Embryonic stem cell transcription factor Sox2 in lung squamous cell carcinoma, adenocarcinoma, small cell carcinoma, large cell carcinoma and adjacent lung tissue nucleic acid and protein levels were expressed, and the protein levels and the expression of the nucleic acid level consistent. Sox2 expression in lung tissue was significantly higher than in adjacent lung tissue. Sox2 expression in lung cancer differentiation and clinical stage. The results suggest that the transcription factor Sox2 expression may be associated with lung cancer occurrence and development. Embryonic stem cell transcription factor Sox2 will likely early diagnosis of lung cancer as a valuable new molecular markers, and to help determine the occurrence and development of lung cancer.
