Abstract


Objective animal experiments and investigation of occupational groups, to clarify the mechanism of The bromopropane Two isomers (male) reproductive system toxicity, and to explore bromo propane reproductive toxicity in animal models. In this study, eventually shed new the bromopropane human reproductive toxicity, and to develop the bromopropane the occupational health standards provide a theoretical basis. Method 1 select certain provinces of 5 1 - bromopropane production plant 85 male workers exposed group, 86 male workers without exposure to toxic and hazardous chemicals for the control group. Trained professional investigators who conducted a questionnaire survey on two groups of workers, using gas chromatography mass spectrometry analysis of workers' exposure to the outside of bromopropane exposure, and workers collected semen routine analysis. 2.21 male rats were randomly divided into control group, 1 - bromopropane group and 2 - bromopropane group the bromopropane treated daily by intraperitoneal injection 1g/kg bromopropane, control group was injected solvent olive oil, intraperitoneal injection 2 weeks. Of rat epididymal sperm count and morphology evaluation; using HE staining evaluation of testicular pathological changes; using TUNEL staining and Caspase-3 activity immunohistochemical method to evaluate apoptosis in rat testis damage; using chemical colorimetric determination of the epididymis and testicular GSH and MDA oxidative stress indicators; quantitative real-time PCR and Western blotting of apoptosis-related genes Bax, Bcl-2, p53, Fas and FasL mRNA and protein expression levels determination. Results of a population survey, 1-BP low exposure group of workers semen quality compared with the control group no significant changes in population exposed to 1-BP group and the high exposure group of workers viscosity of semen and sperm was significantly lower than the control group ( P lt; 0.05). The results of animal experiments, 1-BP exposure of the body weight of rats and genital weight were lower than the control group (P lt; 0.05), and-BP affects only the body weight of rats; compared with the control group, BPs exposure so absolute number and relative number of rat epididymal sperm were significantly lower (P lt; 0.01), 1-BP sperm the tailless increase in the proportion of (P lt; 0.01), and 2-BP mainly in the rat sperm head abnormalities The increase in the proportion of sperm (P lt; 0.01); the control group and 1-BP testis biopsy no obvious abnormal changes in the seminiferous tubules and germ cells, and 2-BP exposure of the rat testis germ cell seminiferous tubule atrophy necrosis, and the emergence of vacuoles; The TUNEL results show that rare positive apoptotic cells in the control group rat testis seminiferous tubules BPs so that the average of the rat testis per seminiferous tubule positive cells was significantly increased, the difference was statistically significant (P lt; 0.05) compared with 1-BP and 2-BP apoptosis serious; 2-BP exposure makes Caspase-3 active activity was significantly higher than that in control group. Compared with the control group, 1-BP rats testicular tissue changes in mRNA and protein expression levels of apoptosis-related genes was no difference, and 2-BP exposure of rat testis can p53, Bax and Bcl-2 genes down mRNA and protein expression levels were statistically significant (P lt; 0.01, P lt; 0.01, P lt; 0.01), but the change of Fas and FasL compared with the control group, the difference was not statistically significant. Conclusion The study found that 1-BP production factory workers, 1-BP workers in contact with a certain degree of reproductive toxicity. The results of animal experiments also confirmed that under the conditions of the same dose of BPs two isomers are rats having a certain toxic effect, but both having the type of the role of the target organ, and 2-BP reproductive toxicity may be higher 1-BP. 1-BP may affect the sperm maturation, the capacitation or other means, cause sperm release delay and abnormal sperm morphology. 2-BP may be causing germ cell degeneration and death through the activation of germ cell apoptosis, reproductive toxic effects.
