Abstract


The the of heme oxidase of Ⅰ (heme oxygenasel, Hoxl) and phycocyanin ferredoxin reductase (ferredoxin oxidoreductase, PcyA) is a key enzyme in the synthesis process of phycocyanin choline (PCB) , they express two enzymes the catalytic heme (heme) into choline phycocyanin genes control the synthesis of these two enzymes are of of heme oxidase I gene (hoxl) and phycocyanin ferredoxin protein reductase gene ( pcyA ) . The phycocyanin crack together the isomerase, E and F (cpcE / F) is responsible for the catalytic PCB apoprotein connection . Only a combination of color -based phycocyanin to optically active . With the the phycocyanin is more and more widely used in the production life of about hoxl Genes and pcyA of gene cloning and sequencing features analysis become a the phycocyanin hot topic . The experimental application touchdown PCR method amplified from SYNECHOCOCCUS the Synechococcus cedrorum of the 717bp of hoxl gene sequences , and compared to the to download 13 of cyanobacteria hoxl gene sequences from GenBank using MEGA4.0 sequence homology analysis and protein structure characteristics analysis . The results show that , Synechococcus sp. Cedrorum the hoxl gene , Synechococcus Synechococcus PCC7942 and S.platensis , section spin algae Arthrospira platensis hoxl gene sequence homology were 99.9% and 99.2% amino acid sequence homology of 99.6% and 97.9% , respectively . Also build a system using the neighbor-joining and maximum parsimony tree and cluster analysis . The use of PCR technology from the the algae algae , Spirulina Arthrospira strains of Arthrospira platensis FACHB314 cloned 747bp of pcyA genome sequence and compare to download three of cyanobacteria pcyA gene sequences from GenBank sequence homology analysis and protein structure with MEGA4.0 characterization . The results show that , The Arthrospira the platensis Discovered in FACHB314 pcyA gene Synechocystis Synechocystis sp. PCC 6803 the highest similarity of 99.9% and 99.6% amino acid sequence homology . Use the In-FusionTM kit to construct a recombinant plasmid pUC18-HP plasmid pSB130-HP , the PCR method and double digestion results show that : the expression vector pUC18-HP the plasmid and pSB130-HP plasmid was constructed successfully .
