Abstract


The near-infrared spectroscopy technology is one of the fastest detection technology developed in recent years , with a non-destructive , rapid, efficient , convenient and environmentally friendly features . In the logarithmic growth phase of E. coli and Listeria monocytogenes bacteria as the research object , examines the near-infrared light on the mechanism of action of the pathogen cells , pretreatment methods of Near Infrared Spectroscopy projection discriminant analysis based on principal component analysis techniques for the identification of pathogenic bacteria , to get the desired results . 1 photoelectric turbidimetry and plate count method , the logarithmic growth phase of E. coli . Compare the growth of E. coli graph to study its growth trend , identified by the logarithmic growth phase of E. coli , the last selected 14h incubation time is Escherichia coli . By single factor and orthogonal experiments , crushing parameters of E. coli cells were studied. The crushing rate as an indicator E. coli cells , E. coli cells broken parameters , determine the optimum crushing combination of parameters : the crushing power 400W, each broken 5s interval 5s, actually broken a total time of 50min . 3 according to the different concentrations of E. coli cell wall and cytoplasm of the near-infrared spectrum , respectively, examine the vector owned by a law , multi- scattering correction method , a first-order guide number method , the first order derivative vector owned by a method such as near-infrared analysis technology of spectral pre- processing technology , the results show that the first derivative method and the first derivative vector normalization method for spectral preprocessing very satisfactory , while the use of vector normalization method and multivariate calibration method for spectral preprocessing satisfactory results can be obtained . 4 . Projection discriminant analysis based on principal component analysis technique to study the near-infrared light on the role of the E. coli cell wall and cytoplasm , results showed that the pathogen concentration of near-infrared light absorption into the concentration ratio in the cell wall and cytoplasm of near-infrared light absorption is significantly different , namely the use of near-infrared absorption spectroscopy can distinguish between the two . 5 projection discriminant analysis based on principal component analysis technique , whether the use of cell wall or cytoplasm near-infrared spectroscopy , can increase the E. coli and single Listeria distinguish , but the cell wall of near-infrared spectroscopy to distinguish two pathogens, higher accuracy .
