Abstract


Carbonyl poisoning theory of aging is exploring age pigment formation mechanism based on the integrated radical theory of aging and glycosylation summative theory of aging theory of aging . This doctrine that free radical damage and non- enzymatic glycation damage formation of biologically toxic intermediates of unsaturated aldehydes and ketones aging is caused by one of the most important reasons . This paper reviews the biochemical mechanism of the formation of age pigment , and is committed to improving age pigment determination and use of new method to detect artificial elderly substances. This dissertation research work are summarized as follows : First, the use of three-dimensional front surface fluorescence and fluorescence spectrophotometry aged mice and young mice rat tail collagen protein in the aqueous phase and an organic extraction liquid substances and residues after extraction fluorescent substance changes. Thus creating age pigment substances measured in vivo content of new methods - dimensional front surface fluorescence method . The results showed that : the first three-dimensional surface of the measured fluorescence techniques age pigment substances content in vivo accumulation over time basically consistent with the extraction test results . This indicates that the front surface of the three-dimensional fluorescence studies on age pigment which will become a very useful tool . Second, from a different rat tissues ( liver, brain , heart, kidney ) and MDA artificial age pigment and biological macromolecules (BSA, DNA) reaction products under the improved methods were tested, experimental results show that they have similar fluorescence properties. Tissue samples after UV irradiation , the fluorescence microscope under different excitation blocks , have issued a similar fluorescent , the sample surface at the same time also the first three-dimensional fluorescence detector . MDA and biological macromolecules (BSA, DNA) fluorescence characteristics of the reaction products is relatively simple , MDA reaction with DNA is only one pair of peaks . Our experimental results show that the different organizations in the carbonyl stress , the resulting end product may have a similar structure.
