The Effect of Hemorrhage on Neural Behavior and Cell Apoptosis in the Hippocampus CA1 Zone of the Rats under Controlled Hypotension

Abstract


Objective: To observe the the controlled hypotension blood loss in varying degrees of neurobehavioral and apoptosis in hippocampal CA1 region, the study of controlled hypotension in rats blood loss tolerance. Methods: 48 SD rats were randomly divided into Group C (not buck Group) and H group (controlled hypotension group) into two groups, each group within the body by blood loss accounted for a percentage of the total blood volume of each divided into 4 groups : C1, H1 group (10%), C2, H2 group (20%), C3, H3 (30% group) and C4, H4 (40% group), n = 6. Group H with sodium nitroprusside (SNP) combined with esmolol (ESM) controlled hypotension to mean arterial pressure (MAP) of 50 to 55mmHg stable after 10 minutes to maintain the speed of the pump and start femoral artery bloodletting, C group infusion of saline, the bloodletting process is the same as Group H after 60 minutes re-pressed, recording vital signs and intraoperative monitoring of arterial blood gas. Behavioral changes observed after 24 hours survival rat open field activity score and the inclined plane test, detected by immunohistochemistry detection area apoptosis of hippocampal CA1 region of bcl-2 and caspase-3 expression in situ end labeling technique (TUNEL method) cells, calculated 24h survival rate. Results: 24h survival rate of C1, C2, C3 group were 100% the C4 group for 50%, H1, H2 group was 100%, H3, H4 group were 83% and 17% of the comparison group H4 and H3, H2, H1 group were significantly different (P <0.05), the same amount of blood loss between the two groups was no significant difference (P> 0.05). Survivors open field activity score and the inclined plane test group and the same amount of blood loss between the two groups showed no significant difference (P> 0.05). bcl-2 and comparison group H3 average optical density of caspase-3 group compared with H1, H2 group were significantly different (P <0.05), the same amount of blood loss between the two groups compared with H3 group than in the C3 group increased (P <0.01 and P <0.05). The TUNEL assay percentage of apoptotic cells (AI) group compared with H3 group compared with H1, H2 group were significantly different (P <0.05), the same amount of blood loss between the two groups compared with H3 group than in the the C3 group increased (P <0.05). Conclusion: pure blood loss by 10%, 20%, 30% of the rats after 60min recovery 24h after reperfusion did not cause the occurrence of apoptosis of hippocampal CA1 region. SNP composite ESM controlled hypotension, blood loss by 10% and 20% to maintain 24h 60min reperfusion hippocampal CA1 region apoptosis, but the blood loss of 30% of the hippocampal CA1 region of Bcl-2, caspase-3 expression increased apoptosis cells increased, that increase the 30% of the rats with hemorrhagic brain damage caused by controlled hypotension. Composite ESM 2.SNP controlled hypotension in rats under varying degrees of blood loss during early reperfusion after neurobehavioral no difference. Composite ESM 3.SNP controlled hypotension in rats can not tolerate blood loss by 40%, the survival rate is low.
