Abstract


Objective To explore the effective method of in vitro induced rabbit bone marrow mesenchymal stem cells to smooth muscle cells , in order to provide a stable source of seed cells for subsequent experimental work . Bone marrow mesenchymal stem cells obtained from rabbit bone marrow fluid density gradient centrifugation in primary and subcultured observed morphological changes and identified . Induced culture supernatant 5 - azacytidine , smooth muscle cells and smooth muscle cells co-cultured induced induced to smooth muscle cells , the induction efficiency of various methods of induction by morphological and immunohistochemical methods . Results of primary cultured rabbit bone marrow mesenchymal stem cell morphology is more uniform , showed a fibroblast-like , rapid growth and proliferation , subculture grew well . Cultured cells by flow cytometry the appraisal , CD34 and CD45 expression is very weak , and the strong expression of CD90 , prove the cultured cells as compared to pure bone marrow mesenchymal stem cells . In each group induction experiments , the highest 5 - azacytidine induction and co-culture induced by combined treatment group induced group efficiency , simple co-culture induced group induction followed by combined treatment group with 5 - azacytidine induction and supernatant induced rate of two groups without significant differences, merely the lowest the supernatant induced group efficiency Conclusion density gradient centrifugation and adherent screening pure bone marrow mesenchymal stem cells can be obtained . 5 - azacytidine induced by co-culture combined treatment effectively induced to smooth muscle cells .
